Objectives:The aims of this study were to compare the plasma levels of PDGF-BB between the control group and different clinical types of coronary heart disease,and the coronary artery disease And rs1800817 locus,rs2285094 locus genotype difference,and to investigate the relationship between plasma levels of PDGF-BB and its gene polymorphism in patients with coronary heart disease,coronary heart disease(CHD)and the severity of coronary artery disease.Methods:1.In this study,a total of 262 patients from May 2014 to Oct 2015 in the Department of Tianjin Chest Hospital:(1)according to the results of the coronary angiography,they were divided into two groups,CHD group(n=176)and control group(n=86);(2)according to patients' history,symptoms,laboratory examination,the CHD group were divided into SAP group(n=57),ACS group(n=119);(3)According to the results of coronary angiography of the number of lesions,the ACS group was divided into single vessel disease group(n=38),double branch disease group(n=35)and multivessel disease group(n=46).2.The basic data were collected from all subjects including gender,age,height,weight,smoking history,hypertension history,diabetes history,family history of coronary heart disease.Taking venous blood of all subjects after fasting for 12 hours,were tested for(TC,TG,LDL-c,HDL-c,hs-CRP),Fasting blood glucose(FPG),WBC,PLT.PDGF-BB level was tested by ELISA.Human blood cell DNA Extraction Kit was used to extracte human genome DNA,and Real-Time Taqman PCR method was used to detect the genotype of rs1800817 locus and rs2285094 locus.3.Comparison with the control group and the coronary heart disease group,the control group and the SAP group,ACS group and between the control group and the coronary artery of general clinical data,laboratory test differences;The plasma PDGF-BB levels were compared between the control group and the coronary heart disease,the different types of coronary heart disease and the different number ofcoronary artery lesions;Analysis of rs1800817 locus,rs2285094 genotype and allele frequency in the control group and the coronary heart disease,the different types of coronary heart disease and the different number of coronary artery lesions in the distribution of the difference;The relationship between different genotypes and plasma levels of PDGF-BB were compared,and the relationship between different genotypes and different types of coronary heart disease,coronary heart disease and the severity of coronary lesions were compared.Results:1.General clinical data and laboratory examination,the coronary heart disease group compared with the control group,the age,gender,family history of coronary heart disease,hypertension,diabetes,smoking history,BMI,FPG,TC,TG,LDL-c,HDL-c,hs-CRP,WBC,PLT showed no significant difference(P > 0.05).The level of hs-CRP in group ACS was significantly higher than that in control group,the difference between SAP group was statistically significant(P < 0.05),and the level of hs-CRP in SAP group was not statistically significant compared with that of control group(P >0.05).Compared with the control group and the number of lesions in the coronary artery lesions,the proportion of hypertension in multivessel disease group was higher than that in the control group,the difference was statistically significant(P < 0.05),and the differences of laboratory indexes were not statistically significant(P>0.05).2.Comparison of PDGF-BB plasma levels: in coronary heart disease group was higher than the control group,the difference was statistically significant(P<0.05);ACS group was higher than control group,SAP group,the difference was statistically significant(P<0.05),there was no significant difference between SAP group and control group(P>0.05);the single vessel group,the two vessel group and multivessel disease group were higher than control group,the difference was statistically significant(P<0.05),the difference between the three groups were not statistically significant(P>0.05).3.SNP gene polymorphism analysis:in the control group and the coronary heart disease group,the different types of coronary artery disease and the different numberof coronary artery lesions,rs1800817,rs2285094 genotype distribution difference was not statistically significant(P>0.05).Comparison of PDGF-BB plasma levels in different genotypes,AA and AC+CC,TTand TC+CC,there was no significant difference in the same group(P>0.05).There was significant difference between the control group and CHD group in rs1800817 locus of the AA genotype(P<0.05);The rs2285094 genotype of TT genotype in CHD group was higher than that of control group(P<0.05).4.Correlation analysis showed that plasma PDGF-BB level was not associated with Gensini score.Logistic regression analysis showed that the high plasma level of PDGF-BB was a risk factor for coronary heart disease,high HDL-c level was a protective factor for coronary heart disease.Conclusions:1.High PDGF-BB plasma levels may be a risk factor for coronary artery disease,and may be associated with the stability of coronary atherosclerotic plaque,but is not associated with the severity of coronary artery disease.2.The AA genotype of rs1800817 locus and the TT genotype of rs2285094 locus are associated with high PDGF-BB plasma levels,suggest that AA genotype and TT genotype may be associated with the incidence of coronary artery disease by affectting plasma levels of PDGF-BB.
