Objective:To explore the significance of expression changes of p190 MetNC in the residual cancer cells of rabbits VX2 liver tumor after radio-frequency ablation(RFA)therapy.Methods:Select 20 New Zealand white rabbits,2 of them as the extending rabbits and the others as experimental rabbits.We planted the VX2 tumor vaccination within the lateral muscle strain broth of the rabbit hind legs.The left hepatic lobe tumors growing demonstrated that VX2 tumor model is set up.Ultrasonic monitoring,after the success of the models,the experimental rabbits were randomly divided into three groups(including group A,group B and group C),and each group has 6 experimental rabbits: group A for the control group,no radio-frequency therapy;group B for single radio-frequency therapy;group C for radio-frequency ablation group again and again.Ultrasound will kill A tumor-burdened rabbit,respectively,after cutting specimens,immune histochemical methods are used to observe group A p190 MetNC expression in tumor tissue and normal tissue,B and C group rf area and residual carcinoma area p190 MetNC expression.Statistical difference analysis.Results:1.The building results: 2 weeks after inoculation,ultrasonic inspection showed 18 experimental rabbits were successful.Ultrasound showed round or class round in the liver low echo tubercle,1.5 cm in diameter,borders,no envelope echo,blood flow signal is rich,and visible spectrum.2.The ultrasound results after RFA :group B after single RFA: postoperative residual carcinoma boundary is not clear,irregular form.4 weeks after postoperative,tumor recurrence,similar size with preoperative,irregular hyperechoic treatment area,the center is a small amount of irregular anechoic no obvious blood flow signal;Group C after repeatedly RFA(3 times),4 weeks later tumor recurrence,the size is increased preoperative,irregular hyperechoic treatment area,central irregular anechoic,residual carcinoma of visible blood flow model.3.HE staining results: the tumors of group A and the residual cancer of groups B and C showede enlarged hyperchromatic nuclei,pleomorphism,distribution in nests.Visible area of groups B and C after RFA showed coagulation necrosis,nuclear pyknosis,karyorrhexis,karyolysis and there were a lot of red dye around organization,lymphocytic infiltration,inflammatory reaction zone formation.4.The immunohistochemical method was used to detect the expression of p190 MetNC in different area of each group.SPSS software statistical analysis: the expression of p190 MetNC in tumor tissues of group A is(8.20±3.2)%,no p190 MetNC expression in normal tissue;The positive expression of p190 MetNC in residual carcinoma of Group B is(15.68±5.73)%,(2.48±1.56)% in radio frequency area;The positive expression of p190 MetNC in residual carcinoma of Group C is(21.70±9.38)%,(2.33±0.59)% in radio frequency area.Conclusion The expression of p190 MetNC in residual tumor cells: RFA therapy can not only destroy tumor tissue and make them necrosis,but also stimulate high expression of p190 MetNC in residual tumor cells.Eventually RFA therapy lead to tumor recurrence.The high degree of expression may be related to the number of RFA therapy.
