Objects:1. To master the method of establish the rabbit VX2 hepato-carcinoma model;2. To study the influence on the animal models by radio-frequency ablation therapy (RFA);3. To explore the apoptotic mechanism of the rabbit VX2 hepato-carcinoma models following RFA and its significance;Methods:1. The 70 expermental hepato-carcinoma models, which were established by injecting VX2 tissue suspension into the rabbit livers, were divided into 2 groups randomly. (1) The experimental group including 35 rabbit models was treated by RFA. They were anesthesized by 846 mixture injecting into muscle and incided through the abdomen median incision, and then the tumors were found and measured. The RFA needle was inserted into the center of the tumor, the radio-frequency (RF) generator was triggered, and then the RFA began. The therapy was finished after 8 1.3min. (2) The control group including 35 rabbit models was treated by pseudo-RFA. The same method was used as the fomer without the RF generator triggered.2. All the experimental animals were killed in Oh, 12h, 1d, 2d, 4d, 8d and 16d respectively after RFA. The ringle and the central tissues of the experimentaland the control group were cutted, fixed by 100ml.L-1 formaldehyde solution, embedded by paraffin, sected, dyed by hematoxylin and eosin (HE), and conducted by TdT-mediated X-dUTP nick end labeling (TUNEL) respectively and sequently.3. The liver function medical index was detected before and after RFA, andtheir consequences were contrasted and compared.4. All the experimental animals were observed by ultrasonography. Results:1. The 70 rabbit hepato-carcinoma animal models were built successfully and confirmed by pathological examination, whose successful rate was 100%.2. After RFA, their appetite decreased apparently, the majority of them were indepression and drowsiness and moved little, and the minority of them were agitated easily. Then they restored to the pre-therapeutic condition after 3~ 4d and their difference diappeared. The animals of the control group showed that their appetite decreased and they moved little after 4 weeks. They would die naturally without therapy after 8 weeks.3. The total protein and the albumin of the rabbit models decreased apparently, and the total bilirubin, the aspartate aminotransferase (GOT), the glutamic-pyruvic transominase (GPT), and the alkaline phosphatase increased. The total protein and the alkaline phosphatase restored to the normal condition for 1 week, but the others restored about 2 weeks.4. The size of the tumor decreased after FRA, and their HE dying consequencesshowed that the apoptotic cells increased.5. A great deal cellular necrosis and more apoptotic cells and bodies wereobserved by transmission electron microscope.6. The apoptotic cells were deteced by TUNEL. A great deal positive cellswere found in the central tissue of the experimental group and more positive cells were also found in the ringle tissue, whose difference was verysignificant (P<0.01) . A little positive cells were found in the control group, and there was no difference between the ringle and the central tissues. The difference between the apoptotic index of the experimental group and the control group was very significant (P<0.01) . The apoptotic peak of the experimental group appeared at the 24 hour time-point. The apoptotic cellular number of the control group did not vary with time. Conclusions:1. RFA is an effective therapeutic method of liver carcinoma with micro-trauma.2. Because RFA can cause bad influence on the liver function, the liver function protection and supporting therapy should be paid more attentions to the clinical process of RFA.3. It is an important mechanism of treating liver malignant tumor by RFA toinduce the apoptotic cells. The main apoptotic spot lies in the ringle region of the tumor and the apoptotic peak appears at the 24 hour time-point.
