Objective:To discuss the expression of the cysteine-containing aspartate-specific proteases (caspase-9) in rabbits liver VX2 tumor-remnant tissues after radio-frequency ablation.Methods:The VX2 tumor cells were implanted on the inside of a hind leg muscles of rabbits,2 weeks after the implantation substantial mass were palpable,that is tumor-bearing rabbits.Cut the fish-like organization at the size of near 1 mm3 pieces, which cut from the edge of tumor in the leg.VX2 tumor were implanted in 19 tablets under ultrasonography guidance.Models of rabbit VX2 liver tumor were set up. A random sample of two death with HE staining were observed under microscopic to determine whether the tumor inoculation successfully.Models of rabbit VX2 liver tumor were divided 4 groups at random. There are 2 rabbits in group A,as the control group, no RFA treatment.Every rabbits in B,C,Dgroup were subjected to RFA treatment. Tumor-remnant tissues were remained by controlling damage scope at 2/3 of the largest radius of the tumor.15 rabbits were divided 4 groups According to their different time after treatment.That is after RFA group (Bgroup),one week after RFA group (Cgroup), two week after RFA group (Dgroup).There are 5 rabbits in every group.After observation of tumor-bearing rabbits were executed,The expression status of caspase-9 in rabbit liver VX2 tumor-remnant tissues was detected after radio-frequency ablation treatment with immuno-histochemical techniques.Results:1.The ultrasonographic results showed the round or circular hypoechoic nodules,borders clear,echo-free capsule,rich blood flow signals in liver and can explore and arterial blood flow at 21 d after inoculation.HE staining cells showed nested distribution,large size,irregular,large and deeply stained nuclei,showing the success of liver implantation.2.Postoperative RFA ultrasound showed high echo mass with irregular border was not clear,irregular shape at tumor destruction zone.CDF and CDE can not detect blood flow signals.3.Immunohistochemical results:Before radio-frequency ablation, Cas-pase-9 in rabbit liver VX2 tumor-adjacent tissues was in low level express-ion, with expression rate 9.6±5.3%.After the radio-frequency ablation, the expression rate of caspase-9 in tumor-remnants tissues was 13.6±9.8%.One week after the radio-frequency ablation,the expression rate of caspase-9 in tumor-adjacent tissues was 46.8±12.8%.Two weeks after treatment,the expression rate was 76.7±15.5%.Conclusion:The radio-frequency ablation can effectively eliminate the liver tumor cells in tumor-adjacent tissues.
