Experimental Study On The Effect Of Modified Aconiti Decoction On Serum Cytokines In Rats With Knee Osteoarthritis

Objective : Through the observation of modified Aconiti Decoction on knee osteoarthritis model in rats serum cytokine interleukin-1 beta(IL-1?),tumor necrosis factor alpha(TNF-?),matrix metalloproteinase-1(MMP-1)content influence,to explore the relationship between IL-1?,TNF-?,MMP-1 and alpha beta for knee osteoarthritis the study on the mechanism of Jiawei Aconite Decoction in the treatment of knee osteoarthritis,and provide a theoretical basis for its clinical application.Methods : The 40 experimental rats were randomly divided into experimental group(Modified Decoction group),control group(Bi Qi capsule group),model group and blank group(n = 10).In addition to the blank group,the other three groups of rats through the modified Hulth modeling method to replicate the stable rat model of knee osteoarthritis.After 7 days by gavage,the experimental group and the control group were treated by Decoction of Aconite Decoction,Biqi Capsule,model group and blank group were given same volume physiological saline.After 28 days of medication,the abdominal aortic blood sampling blood,serum samples of rats by centrifugal method.Enzyme linked immunosorbent assay(ELISA)was used to detect the concentration of IL-1?,TNF-and MMP-1 in serum samples.Objective To observe the effect of modified Aconiti Decoction on serum cytokines(IL-1?,TNF-?,MMP-1)in model rats.Results : The rat model of knee osteoarthritis was successfully established,the results of serum cytokine detection: Compared with the blank group,the levels of IL-1?,TNF-?and MMP-1 in serum of experimental group and model group were significantly increased,and the difference was statistically significant(p<0.01).Compared with the model group,the serum levels of IL-1?,TNF-?and MMP-1 were significantly decreased in the experimental group(P<0.01).Compared with the control group,the serum levels of IL-1? and TNF-?in the experimental group were significantly reduced(P<0.01),and there was no significant difference in the content of MMP-1(P>0.05).Conclusion : Serum cytokines IL-1 ?,TNF-?,MMP-1 play an active role in the pathogenesis of osteoarthritis.Modified Aconiti decoction can decrease the levels of IL-1?,TNF-? and MMP-1 in the serum of rats with knee osteoarthritis,which related to the mechanism of knee osteoarthritis.
