Diabetic retinopathy is a serious complication of diabetes,patients with serious impact on visual function,is the main cause of blindness in the present work,the blindness rate is high,high incidence rate,has become the main cause of blindness,is now the focus of prevention of blindness.The specific mechanism is not very clear now,in the course of the occurrence of diabetic retinopathy due to hypoxia and other factors lead to increased secretion of vascular endothelial growth factor,the influence of VEGF overexpression of vascular permeability,leading to blood retinal barrier damage,is one of the main pathogenic factors.And the study showed that protective effect of schisandrin B on oxidative stress in liver cells,heart and brain ischemia reperfusion injury,and on whether DR has protective effect has not been reported.In this study,we based on the STZ induced diabetic rats were intervened by SchB,to investigate whether SchB can inhibit retinal VEGF over expression of diabetic rats,delay the progress of DR,and further investigate the mechanism for the treatment of DR,and provide a new way of thinking.ObjectiveThrough the SchB intervention in SD rats induced by STZ,expression of VEGF was detected in SD rats blood retinal barrier integrity and retinal tissue,to investigate the integrity of SchB is to help maintain blood retinal barrier in diabetic rats,and investigate whether SchB can maintain the blood retinal barrier by inhibiting the expression of VEGF and provide new ideas for the treatment of diabetic retinopathy.Method50 male Sprague-Dawley(SD)rats were treated with clean grade male(55-70%)for about 8 weeks,weighing about 250 g.The feeding conditions were as follows: room temperature 18-25 0C,air circulation,relative humidity.Daily cage to keep their living environment is basically dry.During the experiment the rats ad libitum drinking water,feed for the full price in grain and domesticated a week(reared in traditional Chinese Medicine Research Institute of Henan province),except normal control group were made using STZ diabetes model after model were randomly divided into normal group(14 rats)and experimental group(14 rats).The model group(14 rats),the experimental group were given different doses of SchB gavage.After 6 weeks of retinal leakage test and VEGF by enzyme linked immunosorbent assay for detection,to detect the value of statistical analysis.Result1.SchB intervention group,the amount of EB leakage in the retina of diabetic rats(low dose of 5.957± 0.126,high dose of 5.957±0.126)was significantly lower than that of the control group(9.376±0.249),the difference was statistically significant(P<0.001).2.SchB intervention group,the amount of VEGF protein in retina of diabetic rats(low dose of 0.469 ± 0.161,high dose of 0.355± 0.013)was significantly lower than that of the non intervention group(0.526±0.017).Conclusion1.Schisandrin B can inhibit retinal vascular leakage and maintain the stability of blood retinal barrier in diabetic rats;2.Schisandrin B can delay the expression of VEGF and inhibit the progression of diabetic retinopathy.
