Objective:To study the regulation effect of Anshen Dingzhiling(ADL)on levels of monoamine neurotransmitters and their metabolites in prefrontal cortex of spontaneously hypertensive rats(SHR),and reveal the mechanism of ADL in treatment of attention deficit hyperactivity disorder(ADHD).Methods:The 8 Wistar Kyoto rats were used as normal group 1,and 8 SD rats were used as normal group 2.50 SHR rats were randomly divided into model group,atomoxetine(ATX)group(0.045mg/kg),and ADL low,middle,and high dose groups(6.7,13.4,26.7g/kg respectively),n=8 in each group.These rats in various groups received corresponding drugs by ig administration,bid for 4 weeks.Microdialysis was used to collect the brain dialysate from the prefrontal cortex of the rats awake,and the in vitro recovery rate of the probe was measured.The high performance liquid chromatography-electrochemical method(HPLC-ECD)was used to detect the contents of neurotransmitters NE,DA and metabolites DOPAC and HVA in the brain dialysis fluid.Results:(1)The open field test showed that the model group and the drug treatment groups had a significantly longer moving distance and a significantly higher average moving speed than the normal control group 1 and 2 before administration(P?0.05).Compared with the model group,the ATX group and the ADL middle group had significant reduction in moving distance and average moving speed on day7,day21 and day28(P<0.05).(2)The levels of NE,DA,DOPAC and HVA in the brain dialysate in the model group and drug treatment groups before administration were significantly lower compared with the normal group 1(P<0.05)and the levels of NE and HVA were significantly lower compared with the normal group2(P<0.05).The levels of NE and DA in the dialysis solution of the ATX group after 2 weeks of treatment were significantly higher than the model group(P?0.05).After 4 weeks of treatment,the levels of NE and DA in brain dialysate of the ATX group and ADL middle group were significantly higher than the model group(P?0.05)and the level of DOPAC in the ADL middle group was significantly higher than the model group(P?0.05).Conclusion:(1)Neurotransmitters NE,DA and metabolites DOPAC and HVA are deficient in the prefrontal cortex of SHR rats.(2)ATX and ADL can regulate hyperactive and impulsive behavior of SHR rats and improve low levels of the neurotransmitters,and there is no significant difference between them but ATX may work in less time.(3)ADL may achieve the purpose of treating ADHD by increasing the content of NE and DA in the prefrontal cortex.
