Osteosarcoma is a kind of malignant tumor.It mainly originates from the skeletal mesenchymal cells and belongs to highly malignant bone tumors.It is a common occurrence in adolescents.It is characterized by the continuous differentiation of tumor cells that have been continuously proliferated into bone-like tissues.The occurrence of hematogenous metastasis is early and has a high incidence,so it progresses rapidly and the mortality rate is extremely high.In recent years,the diagnosis of osteosarcoma mainly depends on X-ray film,chest X-ray film,CT and MRI,but all show limitations,inaccuracies,lag,and has a high rate of misdiagnosis.The treatment is currently mainly through surgical treatment and subsequent drug chemotherapy,radiation therapy,interventional therapy,stem cell therapy and so on.However,there are more or less problems.Such as the limitations of surgery;toxic and side effects of chemotherapy;insensitivity to radiotherapy;the difficulty of interventional treatment techniques;the uncertainty of stem cell effects are currently the problem to be solved.Gene therapy is the introduction of exogenous genes(therapeutic genes)into the tumor cells at the gene level to achieve the purpose of treating tumors.This is the reason that the proto-oncogenes are re-inhibited after therapeutic activation to achieve therapeutic goals.This method is considered to be able to solve the proliferation and spread of tumor cells from "this",and is no longer the treatment of "disease" by traditional methods.Therefore,the current exploration and demand for gene therapy is extremely urgent.The aim of this study was to investigate the expression of Livin and p53 in osteosarcoma and the correlation between them.This study provides a theoretical basis for the diagnosis and gene therapy of osteosarcoma and has some clinical significance.Objective1.To study the expression of Livin protein and p53 protein in osteosarcoma;2.The correlation between Livin protein and p53 protein expression in osteo-sarcoma was analyzed.3.Explore the biological mechanism of Livin,provide a reference for the diag-nosis and treatment of osteosarcoma.MethodsA total of 40 cases of osteosarcoma tissue wax blocks from 2005 to 2015 were collected as an experimental group.In addition,20 cases of paraffin blocks were selected as benign control groups.Immunohistochemistry SP method was used to detect the expression of Livin and p53 in chondroma and osteosarcoma,respectively.In addition,the relationship between the two proteins and clinicopathological parameters of osteosarcoma and the correlation of the expression of the two proteins in osteosarcoma were further analyzed.Results1.Expression of livin protein in osteosarcoma tissue and chondroma tissueThe positive expression rate of Livin protein in 40 cases of osteosarcoma was 62.5%(25/40),while it was not expressed in 20 cases of chondroma.The difference between the two groups was statistically significant(χ2=21.429,P<0.01),Livin is highly expressed in osteosarcoma.The positive expression of Livin protein in osteosarcoma was correlated with pathological grade(high,moderate,poorly differentiated)and Ennking stage(Ⅰ-ⅡA,ⅡB-Ⅲ)(χ2=7.138,P=0,028;χ2=6.222,P=0.013),and the age,gender,and osteosarcoma patient’s age.There was no relationship between the location of the tumor and its pathological type(P>0.05).2.Expression of p53 protein in osteosarcoma and chondroma tissues The positive expression rate of p53 protein in 40 osteosarcoma sections was 52.5%(21/40),while the positive expression rate in 20 cases of chondroma tumors was 5%(1/20).The difference was statistically significant(χ2=12.955,P<0.01),ie p53 was highly expressed in osteosarcoma.The positive expression of p53 in osteosarcoma was correlated with histological grade(high,moderate and poor differentiation)(χ2=8.190,P=0.017).It was not related to the age,sex,tumor location,Ennking stage,and histological type of osteosarcoma(P>0.05).3.The correlation between Livin and p53 expression in osteosarcoma tissues.Statistical analysis showed that there was a significant positive correlation between Livin and p53 expression in osteosarcoma.Conclusion1.Livin protein was highly expressed in osteosarcoma and was not expressed in chondroma.The positive expression of Livin was significantly correlated with the differentiation and Ennking stage of osteosarcoma,and the positive rate of Livin was associated with pathological grade and surgical staging.The decreasing trend of the decrease suggests that Livin can be used as a test indicator for the malignancy and therapeutic judgment of osteosarcoma.2.p53 is highly expressed in osteosarcoma and is low expressed in chondroma.The expression of p53 has a significant correlation with the degree of differentiation of osteosarcoma.With the decrease of pathological grade,the degree of malignancy,p53 is positive.The rate of expression also gradually decreased.It suggests that p53 is associated with the degree of malignancy of the tumor and may be used as a marker for the diagnosis,treatment,and prognosis of osteosarcoma.3.Livin and p53 have significant correlations in the expression of osteosarcoma.There is a positive correlation between Livin and p53,indicating that they play an important role in promoting the occurrence and development of’ osteosarcoma.The combined monitoring of Livin and p53 has a certain value in the diagnosis of osteosarcoma and gene target therapy.
