Objective :Oral squamous cell carcinoma (OSCC) are one of the most common malignant tumor in oral and maxillofacial region, One of squamous cell carcinoma of oral tongue cancer is the most common, and in recent years the incidence of gradual upward trend. Although radiotherapy and chemotherapy in recent years made great progress in technology, the survival rate of patients did not significantly improve. Survival in patients with morphological changes and different degrees of loss of function usually exist and some cured patients will relapse in a few years. Therefore, effective treatment for tongue cancer find the target to become the research hotspot in recentyears. nhibitor of apoptosis protein(IAP) family is first found by Losis Miller. etc, who found it in baculovirus group (CpGC) and Orgyiapseudotsugata multinucleusd virus(OPMNV). It is a new kind of anti-apoptotic protein which is independent of Bcl-2. Each contains one of more baculovirus IAP repeat domains, which are necessary to bind specifically to aterminal effector cell-death protease (for instance, caspases-3 and -7). Binding results substantially reduced caspase activity and reduced cell death in response to diverseapoptotic stimuli. Livin found recently was a novel ,inhibitor of apoptosis family members. The distribution of Livim had a high selectivity, tissue-specific expression in human fetal tissues and nearly all solid malignancies. There were little expression of Livin in normal adult tissues. It has been suggested that suppression of apoptosis may contribute to the development and progression of cancer. Anti-apoptotic Livin gene, like suvivin, is highly expressed in cancer cells and transformed cells, but shows little or no expression in normal differentiated tissues. However, the current literature has yet to see tongue squamous cell carcinoma of the expression of Livin reported. To explore the expression of Livin one of inhibitor of apoptosis protein(IAPs) in tongue squamous cell carcinoma(TSCC),and its correlation with Bcl-2 in TSCC in the experiment by Immunohistochemical method.Materials and Methods:There was a total of 45 paraffin specimens of tongue cancer selected in the pathology laboratory of oral Jiamusi University hospital from January 1998 to June 2007. 10 normal lingual mucosa come from fracture tongue Organization(median age 41.2 years).45 cases of tongue squamous cell carcinoma specimens of patients did not recieve any radiotherapy or chemotherapy, and patients were diagnosed by pathological examination。Immunohistochemical (SABC)was used to detect Livin and Bcl-2 . The results were statistically analyzed by SPSS 13.0 software. high, medium and poorly differentiated group of Livin protein expression use non-parametric Kruskal-wallis test. non-parametric Mann-Whitney U test was used in Normal tongue mucosa and tongue squamous cell carcinoma groups, and other clinicopathological features in tongue squamous cell carcinoma groups Livin protein expression . There was a statistical significance with P<0.05. Livin and Bcl-2 in TSCC as well as their correlation in the occurrence and development of TSCC by Spearman.Result: Positive staining rates of Livin in TSCC and NLM were 68.89%and 10% respectively,and there was a significant statistical difference between them(P<0.01).Overexpressions of Livin were related to the pathological grade stage(P<0.05),and closely to cervical lymphatic metastasis of TSCC(P<0.05).Positive staining rates of Bcl-2 in TSCC and NLM were 60.0%and 0%respectively and there was a significant statistical difference between them (P<0.01). There was apositive relation between them, and the Spearman correlation coefficient r was 0.353,P=0.027. Conclusion:1. There is a high positive expression in tongue squamous cell carcinoma with Livn, which is much higher than in normal tongue mucosa tissues.2.The expression of livin is significantly associated with the degree of cancer differentiation and lymph node metastasis, while not associated with age, sex and TNM classification. 3.There is a high positive expression in tongue squamous cell carcinoma with Bcl-2 however there was no expression in normal tongue mucosa tissues. 4.The expression of bcl-2 is significantly associated with TNM classification and lymph node metastasis, while not associated with age, sex and The degree of cancer differentiation.5. Tongue squamous cell carcinoma in the inhibitor of apoptosis protein Livin and Bcl-2 protein expression was positively correlated.
