| Osteosarcoma originates from osteoblastic mesenchymal cells and is a primary or cartilage tumor characterized by proliferating tumors.The cells directly form bone or bone-like tissue,which is the most common primary malignant tumor in children and young patients,which seriously endangers the health of adolescents.The most common lesions of osteosarcoma are the active distal femur and the metaphysis of the proximal humerus.The short course of disease is rapid and easy to metastasize early.At present,with the application of neoadjuvant chemotherapy combined with surgery and multi-drug,the 5-year survival rate of osteosarcoma patients has increased to 60%-75%,but 30%-40%of patients still have lung metastasis and recurrence,and the prognosis is poor.The 5-year survival rate is only 20%.Moreover,as the survival time of patients prolongs the side effects caused by chemotherapy and related problems such as drug resistance,it is necessary to explore new safer and more effective treatment methods.The mechanism of tumor development is still not very clear.It is generally believed that the destruction of normal cell cycle and inhibition of apoptosis are closely related to tumors.Livin is a newly discovered inhibitor of apoptosis protein,which is low or absent in normal tissue cells and highly expressed in tumor cells.It has been confirmed in many studies to be closely related to the occurrence and development of various tumors in osteosarcoma.It plays an important role in the occurrence and development.The Bcl-2 plays an important role in the regulation of apoptosis,and Bcl-2 protein is closely related to the occurrence and development of tumors.The main purpose of this study was to explore the expression of Livin protein and Bcl-2 protein in osteosarcoma and the correlation between these two proteins,in order to provide a theoretical basis and guiding significance for the clinical diagnosis and treatment of osteosarcoma.Objective1.To investigate the differential expression of Livin and Bcl-2 in osteochondromaand osteosarcoma.2.Explore the expression of Livin and Bcl-2 proteins in osteosarcoma.3.To explore the relationship between the expression of Livin protein and Bcl-2protein in osteosarcoma and various clinicopathological parameters,and the correlation between them.MethodIn the Department of Pathology of Shenyang Fengtian Hospital and Liaoning Provincial Cancer Hospital,45 cases of osteosarcoma tissue wax blocks,45 cases of osteochondroma tissue wax blocks,osteosarcoma group were experimental group and osteochondral group were control group.The expression of Livin protein and Bcl-2 protein in osteochondroma and osteosarcoma tissues were detected by immunohistochemistry(SP method).To investigate the relationship between the expression of Livin protein and Bcl-2 protein in osteosarcoma and the correlation between them in osteosarcoma.Result1.Expression of livin protein in isoline,osteosarcoma,and osteochondroma.The positive rate of Livin protein in osteosarcoma tissue was 66.7%(30/45),but in osteochondroma tissue Livin protein was not expressed(0/45),and the difference between the two groups of data was statistically significant(x2=45,P<0.01),and Livin was significantly expressed in osteosarcoma.The positive expression of Livin protein in osteosarcoma tissues of different Ennking stages(Ⅰ+ⅡA,ⅡB+Ⅲ)was statistically significant(x2=7.634,P<0.013).The positive expression of Livin protein in osteosarcoma tissues with different pathological grades(highly differentiated,moderately and poorly differentiated)was statistically significant(χ2=1 2.656,P<0.01).2.Expression of Bcl-2 protein in osteosarcoma tissues and chondroma tissues.The positive rate of Bcl-2 protein in osteosarcoma tissue was 62.2%(28/45),in osteochondroma tissue.The positive rate of Bcl-2 protein was 2.2%(1/45),and the difference was statistically significant(x2=35.046,P<0.01).Bcl-2 was significantly expressed in osteosarcoma.The positive expression of Bcl-2 in osteosarcoma tissues with different pathological grades(highly differentiated,moderately and poorly differentiated)was statistically significant(x2=5.678,P=0.017).The positive expression of Bcl-2 in osteosarcoma tissues of different Ennking stages(Ⅰ+ⅡA,ⅡB+Ⅲ)was statistically significant(x2=5.869,P<0.018).3.Correlation analysis between Livin protein and Bcl-2 protein expression in osteosarcoma tissues.Of the 30 patients with positive expression of Livin protein,24 were positive for Bcl-2 and 6 were negative for Bcl-2.Of the 15 patients with negative expression of Livin protein,4 were positive for Bcl-2 and 11 were negative for Bcl-2.Of the 28 Bcl-2 protein-positive sections,24 were positive for Livin protein expression and 4 were negative for expression.Of the 17 Bcl-2 protein negative sections,6 were positive for Livin protein and 11 were negative.Livin was positively correlated with Bcl-2 expression by Spearman rank correlation analysis(r=0.5 1 9,P<0.01).Conclusion1.Livin was significantly expressed in osteosarcoma tissues,and the positive expression rate in osteosarcoma tissues was significantly higher than that in osteochondroma tissues.The expression of Livin is related to the degree of pathological differentiation of osteosarcoma and the clinical stage of Ennking.2.Bcl-2 was significantly expressed in osteosarcoma tissues,and the positive expression rate in osteosarcoma tissues was significantly higher than that in osteochondroma tissues.The expression of Bcl-2 is related to the degree of pathological differentiation of osteosarcoma and Ennking stage.3.There was a positive correlation between the expression of Livin and Bcl-2 in osteosarcoma cells. |
