Experimental Research On Fuzheng Kangfu Mixture Combined With Cisplatin To Inhibit The Transplanted Gastric Cancer

Objective To research the function mechanism of Fuzheng Kangfu Mixture combined with cisplatin in the treatment of the gastric cancer.Methods Fifty-six nude mice were randomly divided into 8 groups as follows:the model control group,the low-dose group of Fuzheng Kangfu Mixture,the middle-dose group of Fuzheng Kangfu Mixture,the high-dose group of Fuzheng Kangfu Mixture,the cisplatin group,the group of cisplatin combined with low-dose Fuzheng Kangfu Mixture,the group of cisplatin combined with middle-dose Fuzheng Kangfu Mixture,and the group of cisplatin combined with high-dose Fuzheng Kangfu Mixture.There were 7 nude mice in each group.The transplanted gastric cancer model of mice was established by subcutaneous injection of gastric cancer SCG7901 cell line.The Cisplatin as well as Fuzheng Kangfu Mixture were administered drugs continuously for 7 days.The weight and tumor weight were measured and the tumor inhibition rate was calculated.The ELISA method was applied to detect Serum renal function(BUN.Cr)in nude mice as well as the immunomodulator content of TNF-??IFN-??IL-2?IL-6;in addition,PCR was applied to detect the mRNA expressions of the apoptosis target of Bcl-2,Bax and Survivin as well as the Vascular Endothelial Growth Factor(VEGF)in nude mice,Results In comparison with the model control group,except for the low-dose group of Fuzheng Kangfu Mixture,the tumor weights of all the other nude,mice were alleviated in different degrees(P<0.05).Among these,the tumor inhibition rate of the high-dose group of Cisplatin combined with Fuzheng Kangfu Mixture was the highest(66.67%).In terms of blood and kidney function,in comparison with the model control group,the contents of BUN and Cr in the serum of nude mice in the cisplatin group were significantly increased(P<0.01);in comparison with the cisplatin group,the contents of the BUN and Cr in serum of the nude mice in the middle-dose and high-dose group of Cisplatin combined with Fuzheng Kangfu Mixture were significantly decreased(P<0.01).In terms of immunomodulation,in comparison with the model control group,the contents of TNF-?,IFN-?,IL-6 and IL-2 in serum of nude mice in the cisplatin group were significantly decreased(P<0.01).In comparison with the cisplatin group,the contents of TNF-?,IFN-?,IL-2,and IL-6 in serum of nude mice in all groups of cisplatin combined with Fuzheng Kangfu Mixture were significantly increased(P<0.05).In terms of cell inhibition,Fuzheng Kangfu Mixture combined with cisplatin could significantly enhance the expression of Bax,and reduce the expressions of Bcl-2,Survivin,and Vascular Endothelial Growth Factor(VEGF)(P<0.05).Conclusion Fuzheng Kangfu Mixture can enhance the inhibitory effect of cisplatin on the transplanted gastric cancer in nude mice,reduce the effect of cisplatin on blood and kidney function,improve immune function,promote apoptosis of tumor cells,and inhibit the formation of microvessel so as to play the role of inhibiting the tumor growth.
