Objective: To explore the effects of L-carnitine and aerobic exercise on myocardial fatty acid metabolism in overweight mice through the change of the free fatty acid(FFA)?heart fatty acid binding protein(H-FABP)?peroxisome proliferator-activated receptor ?(PPAR?).Methods: After a week adaptive feed in C57 BL / 6 mice, randomly selected 9 from 45 mice only fed chow(NC group),another 36 fed a high-fat diet group(H group),nutritional overweight model was seted up after three weeks of feeding.Then mice in H group were randomly divided into four groups, each group of nine: overweight quiet control group(OC),overweight added L-carnitine group(OL),overweight movement controls group(OE) and overweight movement combined with L- carnitine supplementation group(OEL).OE and OEL group swimming training for 6 weeks, 6 days per week;OL and OEL supply rat chow containing carnitine 1 hour before training every day. Monitoring the change of weight in mice every week during experiment, rats were thereafter decapitated quickly after training,then take out the heart, saved into the refrigerator at- 80 degrees Celsius. Determined PPAR? and H- FABP protein expression in myocardial of overweight mice by using Western Blot imprinting method and the content of alkali in heart muscle and FFA by using enzyme-linked immunosorbent assay and colorimetric method respectively.Results:(1) Compared with the OC group, the content of carnitine in the myocardium in OEL and OL group of overweight mice increased significantly(P < 0.05), but the difference between two groups beyond the statistically significant(P > 0.05).(2) Compared with the OC group, FFA levels in overweight mice myocardial decreased significantly in OEL and OE group(P < 0.05);and the content of FFA in myocardial in OEL group significantly below OE groups(P < 0.05).(3) Compared with the OC group, overweight mice's PPAR? amount of protein expression in myocardium were significantly increased in other three groups(P < 0.05); and OEL group had a significantly higher lever than OE group(P < 0.05).(4) Compared with the OC group, the content of H- FABP protein expression in myocardial in OEL and OE group of overweight mice increased significantly(P < 0.05), but the difference between two groups beyond the statistically significant(P > 0.05).Conclusions:(1)Aerobic exercise can increase PPAR? and H- FABP protein expression level in overweight mice's myocardial,promote the use of FFA overweight mice's myocardial.(2)Aerobic exercise can regulate overweight mice in myocardial H- FABP protein expression, the quantity of added L- carnitine are has not obvious control effect.(3)Combined with carnitine and simple motion can reduce overweight mice in myocardial FFA levels, and sports combined with carnitine effect is better, the mechanism is the movement combined with carnitine can further increase the amount of myocardial PPAR? protein expression in mice.
