Purpose:To explore the movement before and after the produced by intraperitoneal injection of methane saline one-time to male SD rats,the influence of oxidative stress injury after exhaustion exercise,for the application of methane saline in the field of sports science to provide experimental basis.Methods:Twenty-one healthy male SD rats,2 months,weight 200 ~ 250 g,after adaptive feed 1week,were equally divided into to three groups at random: Treated-placebo group(TP),Treated-methane group(TM),Preventive & treated-methane group(PTM).Three groups of rats for 10 min speed of 15 m/min the adaptability of treadmill training,the rest two days later,PTM group rats 5 min before the movement according to the dose of 10 ml/kg body weight methane produced by intraperitoneal injection of saline,TP,TM group not to interfere in rats.Three groups simultaneously run slope of 0 °,the speed of 35 m/min one-time exhaustive treadmill exercise.Determine the exhaustion after the record of the movement time and immediately give the TM,PTM group according to the dose of 10 ml/kg body weight methane produced by intraperitoneal injection of saline,TP group of rats with the same dose and method of the saline treatment intervention.After the end of exhaustion exercise10 min and 3 h respectively to the eye and abdominal aortic vein blood of inner canthus,detection of blood routine,blood glucose and lactic acid concentration,the rest of the whole blood in heparin anticoagulant tube blending,separation of plasma,blood urea nitrogen,total antioxidant concentration and creatine kinase,cereal third transaminase,aspertate aminotransferase,superoxide dismutase(sod)and myeloperoxidase activity.All data was analyzed with SPSS17.0.The experimental results with p < 0.05 showed statistically significant difference.Results:(1)Compared with TP group and TM group,it was found that the treadmill running time was significantly longer(*p<0.05,#p<0.05)in rats treated with methane saline;(2)compared with the TP group,at 10 min after the end of the exercise TM group,the BPC,PCT rised(**p<0.01);at 3h after the end of the exercise,RBC rised(*p<0.05).Compared with TM group,at 10 min after the end of the exercise PTM group,the WBC,Hb reduced(#p<0.05),at 3h after the end of the exercise,the WBC,RBC,HCT,Hb reduced;(3)at 10 min and 3h after the end of the exercise,three groups of ratsblood glucose,blood lactic acid,urea nitrogen had no statistical difference.TM group rats blood CK compared with TP,PTM group,had no difference at 10 min,but significantly increased(*p<0.05,##p<0.01)at 3h;(4)blood ALT of three groups rats at10 min and 3h after the end of the exercise had no statistical difference,while the TM group rats blood aspertate aminotransferase compared with TP,PTM group,had no difference at 10 min,significantly increased(*p<0.05,#p<0.05)at 3h;(5)TM group rats blood myeloperoxidase at10 min is significantly higher(**p<0.01)compared with TP group,and at 3h after the end of the exercise were significantly higher(**p<0.01,##p<0.01)than that of TP,PTM group;(6)at 10 min after the end of the exercise,MDA,SOD and total antioxidant of PTM group were significantly lower(*p<0.05,#p<0.05)than TP group,the TM group;at 3h after the end of the exercise,SOD of the TM group rats significantly increased(**p<0.01)compared with the TP group,total antioxidants has a tendency to rise,SOD of PTM group the significantly increase(**p<0.01)than that of TP group rats,the total antioxidant significantly increased(**p<0.01,#p<0.05)compared with TP and TM group.Conclusions:(1)Before exercise intraperitoneal injection of methane saline can extend the male SD rats exercise to exhaustion time.(2)After exercise intraperitoneal injection of methane saline solution may affect the recovery of the function of the cell membrane,at the same time also to some extent reduce the antioxidant capacity of the rats.(3)Before and after exercise intraperitoneal injection of methane saline solution to some extent can reduce the total antioxidant capacity of the rats.
