[Objective] To compare the effect degree of Phellinus linteuse Polysaccharideextracts with exhaustive exercise on the antioxidant capacity,balance of cytokinesproliferation and differentiation,and explore its potential mechanism.[Method]1. Explore the pharmaceutical performance of Phellinus linteuse PolysaccharideTwenty-eight male KM mice (the average weight was23.05±3.22g)were randomlydivided into four groups(each group with7male mice) according to their weight.Thefour groups were Phellinus linteuse Polysaccharide group(PL)、Tobacco smokeexposure(S)、Phellinus linteuse polysaccharide with tobacco smoke exposure group(PLS)and normal control group(C).Phellinus linteuse polysaccharide supplementsgroups were treated with0.18ml/kg irrigation for4weeks.Tobacco smoke exposuregroups were treated Lushan cigarette in which was produced jiangxi.Normal controlgroup were feed with0.9%NaCL.After KM mice were intervened for4weeksconsecutively,mice heart blood serum was separated,The biochemical parameters ofantioxidant capacity testing (SOD,GSH,CAT, GSH-PX)were evaluated usingbiochemical detection;IL-6in pulmonary tissue were evaluated according to assaykits.2.Explore the changes antioxidant capacity and immunity Under the action ofPhellinus linteuse polysaccharide and exhaustive exerciseFourty healthy male Sprague–Dawley mice were randomly divided into5groups(each group with8male mice),Exhaustive Exercise group(E), Phellinus linteusePolysaccharides group(PL),Phellinus linteuse Polysaccharides with ExhaustiveExercise group(PLE),Physiological Saline group(S) and the normal control group(C).Phellinus linteuse polysaccharide supplements groups were treated with0.3mg/kgirrigation for4weeks.Healthy mice were trained by treadmill training for7 weeks(Treadmill training way:incremental-load exhaustive exercise from15m/min to34m/min with a gentle five-degree slope).Serum was separated from heart after8weeks treatment.Experimentalists separated Spleen,thymus and liver et al in time,thencalculated and record relevant index.ABC-Elisa used to detect the differentiation ofcytokines(IFN-γ,IL-2,IL-4, IL-5,CD4+and CD8+) in mice peripheral blood, andcalculated TH1/TH2in mice peripheral blood with cytokines level.Selected samplesof the left liver from each mouse than removed rapidly with hematoxylin andeosin(HE).Tissue biopsies were made the histopathological analysis.[Result]1.The effect of Phellinus linteuse Polysaccharides on KM mice antioxidantcapacity under the oxidative stress condition①Activities of catalase in tissuesActivities of catalase in pulmonary tissues group S was significantly decreasedcompared with the normal group(P<0.05),but group PL was significantly increased(P<0.05).Group PLS was significantly decreased compared with groupPL(P<0.05).As with pulmonary tissue’s,catalase in liver tissue of Group S wasdecreased compared with the normal group,but lack statistically significant difference.②Activities of glutathione in tissuesThe activities of glutathione in pulmonary tissueof group PL was up at2.0093times compared with the normal control group(P<0.05),group PL was up at1.61times(P<0.01),but group S was low as66%compared with the normal controlgroup(P<0.01).The group PLS and PL of activities of glutathione in liver weresignificantly decreased compared with the group C(P<0.05),other groups have nostatistically significant difference.③Activities of glutathione peroxidase in liver tissueThere were no statistically significant difference of glutathione peroxidase in livertissues among the groups.④Activities of superoxide dismutase in liver tissuesGroup PL appeared maximum value of superoxide dismutase in liver tissues,and group S had minimum value,but both of group PL and S had statistically significantdifference compared with group C.Group S was significantly decreased comparedwith the group PL(P<0.05).2.The effect of Phellinus linteuse Polysaccharides on KM mice interleukin-6under the oxidative stress conditionPhellinus linteuse Polysaccharides prompted increase in IL-6of group PLcompared with group C(P<0.01),but group S was significantly decreased(P<0.01).Group PLS was at similar levels of group.3.Effect of Phellinus linteuse Polysaccharide extracts with exhaustive exercise onhistology of the liver with HENo typical injury change of liver tissue were observed in group PL,PLE andC,which indicating that after seven weeks,indicating that all the mice are healthyexcept subtle differences among them.Hepatocytes of Group PL appeared moreneutrophils compared with other groups.The changes of group E mainly were edema,hyperemia and inflammatory cell infiltration.4.Influence of Phellinus linteuse Polysaccharides with exhaustive exercise on thebalance of Helper T Cells proliferation and differentiation①Interferon-γThe level of interferon-γ in plasma of group PLE was significantly increasedcompared with the normal group(P<0.05),but group PLE was significantly decreasedcompared with the group PL(P<0.05).②Interleukin-2Interleukin-2of group E was significantly increased compared with the normalgroup(P<0.05),mean of the group E was aslo greater than group PL(P<0.05).GroupPLE was at similar levels of group C and had no statistically significant difference.③Interleukin-4Interleukin-4of group E was significantly increased compared with the normalgroup(P<0.05).Mean of the group E was aslo greater than group PL but had nostatistically significant difference.Interleukin-4of group PLE was significantlyincreased compared with the group PL(P<0.05). ④Interleukin-5Group E appeared maximum value of Interleukin-5in plasma,and had statisticallysignificant difference compared with the group C(P<0.05).Interleukin-4of group PLand group PLE was significantly decreased compared with the normal group(P<0.05).⑤CD4+and CD8+countThere were no statistically significant difference of CD4+and CD8+count inplasma among the groups.[Conclusion]1. Phellinus linteuse Polysaccharide extracts can enhance antioxidant abilities of miceunder oxidative stress condition,include boosting activity and number of manyantioxidase,which indicating Phellinus linteuse Polysaccharide extracts can assistbody tissues in cleaning free radical aviod organism injury.2.Phellinus linteuse Polysaccharide extracts can raise immunity of normal controlmice and improve immunosuppression of oxidative stress mice.3. Exhaustive exercise perturb balance of helper T cells proliferation anddifferentiation,cause dominant response of Th2.Unlike the other studys, there is asubdominant response of Th1in this study replaced weak reactiong of Th1Underexhaustive exercise.This result indicated that Phellinus linteuse Polysaccharideextracts can recovery the unbalance of Th1/Th2which cause by exhaustiveexercise,but it can not significantly break Th1/Th2balance of healthy mouse.4. Histology of the liver with HE in light microscopic show that exhaustive exercisecause inflammation,which indicating exercise-induced immunosuppression.Phellinuslinteuse Polysaccharide extracts can repair the damage of hyperemia andinflammatory cell infiltration of liver.It also can improve normal immune functionof normal mice by controlling number of neutrophils in liver.
