Through the collection,identification and evaluation of germplasm resources of Lonicera Japonica Flos and Lonicera Flos,this paper established the identification technology of Lonicera Japonica Flos and Lonicera Flos based on ITS(Internal Transcribed Spacer)sequences,determined the chlorogenic acid content and luteolin content of germplasm resources.It provides the basis for the foundation of Lonicera Japonica Flos and Lonicera Flos industry development in Lishui,Zhejiang.The main findings are as follows:In this study,21 Lonicera Japonica Flos and Lonicera Flos’ s germplasm resources in 9 provinces were collected.Through morphological identification,in addition to Hunan germplasm(Lonicera macranthoides Hand.-Mazz.),the plants of other germplasm are Lonicera japonica Thunb..However,there were obvious variations in long-term natural selection and artificial selection.The cultivated germplasms were obviously differentiated into two types,white flowers and safflower.The flowering period and blade size of wild Lonicera japonica Thunb.was’ t consistent.As the Lonicera Japonica Flos mainly to dry buds medicine,and buds in the dry color and size have undergone a certain change,it is difficult to judge the source plants by morphological observation.By studying the correlation between ITS sequence and morphological traits of 21 Lonicera Japonica Flos and Lonicera Flos’ s germplasm resources in 9 provinces,Lonicera Japonica Flos and Lonicera Flos can will be distinguished by the genetic relationship tree of ITS sequence.At the same time,the mutant safflower cultivars and other cultivars were distinguished,and the identification technology of Lonicera Japonica Flos and Lonicera Flos based on ITS sequence was established.The content of chlorogenic acid and luteolin in 20 germplasms were determined by HPLC(High Performance Liquid Chromatography).The results showed that the content in different germplasms varied greatly.18 Lonicera Japonica Flos’ s germplasms were planted in Lishui.Only 10 of them met the chlorogenic acid content(≥1.5%)and luteolin content(≥0.05%),the qualified rate of only 55.5%.However,the main unqualified germplasms is wild germplasm and Variety safflower germplasm.It indicate that artificial cultivars are better,the introduction of wild germplasm and Variety safflower germplasm need to be cautious.
