Cistanche deserticola Y.C.Ma is a traditional Chinese herbal medicine for replenishing kidney and yang,referred to as Cistanche for short,modern pharmacological studies have shown that Cistanche can enhance immune function,anti-aging and anti-oxidation,anti-fatigue,liver protection,kidney yang,neuroprotection,laxative and other aspects;chemical composition mainly include phenylethanoid glycosides,wood Lactin and glycosides,iridoids and their glycosides,alkaloids,polysaccharides,etc;Current domestic and international research demonstrate,the interactions of Cistanche's active ingredients have not yet been clearly defined.Therefore,the thesis aims at reserching the effect of Cistanche deserticola preliminary,by analysising mainly active ingredients(phenylethanoid glycosides and polysaccharides),and based on antioxidant,anti-fatigue pharmacodynamics experiment study interaction of phenylethanoid glycosides and polysaccharides.1 ReviewIn order to comprehensively examine the quality of desert cistanche and improve the bioavailability of phenylethanoid glycosides and polysaccharides,further clarify the active ingredients of phenylethanoid glycosides and polysaccharides.2 Qualitative analysis of phenylethanoid glycosides and polysaccharides in Cistanche deserticola from different areaCollecting Cistanche deserticola from different areas and prepare 70%ethanol extracts(mainly containing phenylethanoid glycosides)and polysaccharides.It found that the phenylethanoid glycosides in Cistanche desertus were mainly echinacoside and verbascoside,and harvesting in autumn was generally higher than that in spring;the molecular weight distribution of polysaccharides in Cistanche deserticola had also appeared larger Differences;finally,16 polysaccharides from Cistanche deserticola were hydrolyzed,the proportion of glucose reached more than 50%in the tested 7 monosaccharides,followed by galactose and arabinose.3 Study on Antioxidant Activity and Interaction of Phenylethanoid Glycosides and PolysaccharidesAfter purification,the content of Phenylethanoid Glycosides increases to 17%than 70%ethanol extract of Cistanche deserticola,calculated as echinacoside and verbascoside.In vitro free radical scavenging test,the IC50 of phenylethanol glycoside was 14.39?g·mL-1,and the 70%alcohol extract was 44.57 ?g·mL-1;the antioxidant activity was increased by about 2 times.The polysaccharides of Cistanche deserticola were obtained by water extraction and alcohol precipitation method.The polysaccharide content was 86.87%and the IC50 of the antioxidant activity was 261.3 ?g·mL-1,which was only 1/18 of the phenylethanoid glycosides.Thus,phenylethanoid glycosides can be treated as mainly active ingredients for antioxidant effect and the relationship between phenylethanoid glycosides and polysaccharides interactions needs to be explored further.4 Fingerprint-efficacy study of the phenylethanoid glycosides and antioxidant activity of Cistanche deserticolaPharmacological studies have shown that phenylethanoid glycosides have significant antioxidant activity in vivo and in vitro.In this section,the fingerprints of 70%alcohol extracts of Cistanche deserticola were established and test the DPPH scavenging of Cistanche deserticola.analyzing the mainly active ingredients for antioxidation effect by Partial Least Squares analysis,correlation analysis,and gray correlation analysis comprehensively.The results showed that 17 kinds of phenylethanoid glycosides such as echinacoside,verbascoside and its isomer,cistanchein A and its isomers,cistanchein F and its isomers,etc,which all contributed to the scavenging of DPPH varying degrees,and worked by synergistic effect.When the common characteristic peak was assigned,kankanoside F?kankanose?salsaside C1/C2 were identified for the first time in the Cistanche deserticola,and 7 new structural compounds were found at the same time,which provided the database of chemical constituents of Cistanche deserticola.5 Study on Anti-fatigue and Interaction of Phenylethanoid Glycosides and PolysaccharidesPre-experimental results showed that Cistanche deserticola can all significantly extend the exhausted swimming time of mice at 1?2?and 4 g/Kg-1.At Optimal dose 2 g/Kg-1,phenylethanoid glycosides and polysaccharides for mice individually and together,exploring the effect of anti-fatigue under different grouped,the results showed that phenylethanoid glycoside can significantly increase the glycogen content in mice,thereby alleviating fatigue,and polysaccharides have no significant effect on the content of increasing glycogen;while the polysaccharides played a major role in lowering the blood lactate level;final,as for removing the urea content,there was no significant difference in the all group.In a word,phenylethanoid glycosides and polysaccharides play the important role in the anti-fatigue effect through different ways.
