Copper is an important essential trace element in the human body,and plays a variety of physiological roles in the organism.However,excessive copper can cause many diseases,such as Alzheimer's disease,Menkes syndrome,Amyotrophic lateral sclerosis and Wilson's disease.Sensitive and convenient detection and selective removal of excess copper is of great significance for the diagnosis and treatment of these diseases.Among many detective methods for the copper,UV-Vis spectrophotometric method,because of its low cost,fast analysis speed,strong universality,attracted wide attention of scholars.In this paper,a new type of copper ion colorimetric probe was synthesized by the reaction of o-aminophenol and tryptanthrin,and the structure of the probe was characterized.The probe can be rapidly complexed with copper ions to form a complex of 1:1 in the Tris-HCl buffer solution of p H 5.5.A new characteristic absorption peak appears at 623 nm,and the color changes from pale orange to bluish green.After that,a new method of copper ion detection is established.The method can respond specifically to copper ions in the presence of interfering ions,and the detection limit is 16 n M,which is far less than that of WHO(WTO).Based on this,the method is used to analyze the complexing ability of Chinese medicine decoction to copper ions.It is screened out that the 5 flavors of cloves,roses,Rosa chinensis,pomegranate peel and gallnut have strong complexing ability to copper ions,and provide a basis for Chinese medicine to treat copper overload diseases.The complex agents are used for the treatment of copper overload diseases in western medicine.The probe,complexing copper ion in a specific way,may be used for the prevention and treatment of many diseases due to copper ions overload,such as Alzheimer's disease,Wilson's disease,Parkinson's disease and Prion disease.Based on this conjecture,the synthetic conditions were optimized and the solubility was determined.After the synthesis was magnified 40 times,purity and yield can reach 99% and 91%respectively by extending reaction time,recrystallizing and recycling filtrate.
