| Juglans mandshurica is a Juglandaceae,Juglansdeciduous leafarbor,one of the three most valuable hard leaved species,were mainly distributed in the northeast region.Our previous research shows that Juglans mandshurica shell seed also contains a large number of flavonoid active ingredients.In this paper,the Juglans mandshurica shell seeed was used as raw material,and the flavonoids of Juglans mandshurica shell seed were taken as the research object to explore its antibacterial effect and stability,to developed Juglans mandshurica shell seeed flavonoid microcapsules and related products.(1)The flavonoids from Juglans mandshurica shell seed were extracted by chloroform,ethyl acetate and nbutanol.The filter paper method was used to analyze the antibacterial effect of each component.The half-dilution method was used to determine the minimum inhibitory concentration of each component,and its antibacterial mechanism was preliminary explored.The results showed that:The flavonoids of Juglans mandshurica shell had better inhibitory effect on Bacillus cereus and Staphylococcus aureus,but had no obvious effect on Escherichia coli and Salmonella.The main inhibitory components in the purified phase and the chloroform phase,and the purified phase>the chloroform phase;The minimum inhibitory concentration for Staphylococcus aureus and Bacillus cereus was 1.25 and 2.5 mg/mL,and the chloroform phase was 5 mg/mL.Further studies found that the electrical.conductivity of bacteria increased,the total leakage rate increased,leading to an increase in cell membrane permeability.The results showed that the flavonoids from Juglans mandshurica shell seed had inhibition effects on gram-positive bacteria,and the mechanism of action might be to destroy the integrity of cell membrane.(2)Spectrophotometry was used to examine the stability of flavonoids from Juglans mandshurica shell seed.The results showed that:The flavonoids from Juglans mandshurica shell seed were unstable under light,sustained high temperature,strong acid and alkali conditions and the addition of redox agents and metal cations have decreased stability,and CMC concentration has a certain influence on the stability.The results showed that the flavonoids from Juglans mandshurica shell seed should be stored under low temperature,avoid light and weak acid conditions,kept away from redox,Mg2+、Fe+、Fe3+ and high concentration of CMC.(3)The method of complex coacervation was used to select microencapsulated wall materials and the optimum process was determined by orthogonal test.The results showed that:Gelatin and beta-cyclodextrin as wall materials,the proportion of gelatin and beta-cyclodextrin were 1:1(g/g),wall materil concentration of 1%,stiring at 30℃ for 30 minutes,the ratio of core material to wall material were 1:5(g/g).Under these conditions,the microencapsulation of product could reach 76.79%.(4)Based on the above tests,according to the relevant standards,three products of syrups,granules and ’capsules were developed.The results showed that:The relevant indicators of flavonoids from Juglans mandshurica shell seed syrup,granules and capsules comply with the Chinese Pharmacopoeia standards;the flavonoid contents of syrup,granules and capsules were 5.6 mg/mL,47 mg/g and 111 mg/g,respectively;and quality inspection standards was formulated for flavonoid of Juglans mandshurica shell seed granules. |
