Flurbiprofen ester is a mixture with dual-enantiomeric-centers,consisting of four components,and its antifungal mechanism is effectively inhibiting the synthesis of the peripheral double-functional enzyme prostaglandin reductase?cyclooxygenase,cox?in the spine,which reduces the pain of postoperative and cancer.In this study,the adsorption isotherm model of bi-chiral centered drugs was proposed to separate optically pure drugs through a method of simulated moving bed?simulated moving bed,SMB?,offering an essential basis for the commercialization and massive production.It is quite necessary to determine the reliable solute competitive adsorption isotherm.In this paper,the optimum conditions for chiral splitting of the flurbiprofen ester were investigated through a four-element model system,which is a breakthrough compared to the traditional two-element isotherm system.The experimental results show that the Henry constants and selectivity coefficients are successfully obtained by a linear pulse test of the enantiomers.Then,the competitive adsorption isotherm of bi-chiral centered drug chiralcel with non enantiomers in stationary phase Ayh chromatographic column was determined by reverse method.Compared with the determination of the two-part isotherm,as the number of groups increases,the adsorption process of four groups on chromatographic column becomes more complicated,which not only increases the workload of prophase experiment,but also increases the difficulty of late program fitting,so it is necessary to add more genetic algorithm equation to solve the problem in the traditional two-part fitting program.Comparing the theoretical knowledge from several isotherm models,the more model parameters are set up,the greater possibility of success fitting can be achieved.Therefore,we introduce two new parameters directly on the basis of the five-parameter Bi-langmuir adsorption model with the most traditional parameters,which becomes qns,qs,bns,bsa,bsb,bsc,bsd seven parameter Bi-langmuir adsorption model,finally,this study uses seven parameter Bi-langmuir adsorption model,the TD column model,successfully fitting out four group of competitive adsorption isotherm.Additionally,this paper explores the UV-CD-joint experiment of the system,although it is found that the CD response value is not sensitive to the UV-CD,which is not suitable for the online test.However,it opens an alternative way for other online monitoring,such as ORD?optical Rotary dispersion detector?and PL?POLARIMETER?.
