Abalone is one of the great economic value of aquatic products,meat tender,nutritious,high edible and medicinal value.But in abalone processing,will generate a lot of offal and other by-products,these organs are usually discarded,not only a waste of resources but also environmental pollution.Therefore,processing and utilization of abalone viscera it has gradually become the focus of attention to develop abalone resource.In this paper,we uesd abalone viscera as raw matrial to prepare abalone viscera crude extract,abalone viscera protein peptide,abalone viscera polysaccharide by enzymatic hydrolysis.Samples of in vitro free radical scavenging activity,antioxidant activity and enhance immune activity in vivo were studied by us.In vitro free radical scavenging activity experiments showed that:Three abalone visceral hydrolyzates have scavenging activity on DPPH and has a certain dose-effect relationship.Wherein,when the sample at a concentration of 5 mg/mL,enzymatic crude extract of DPPH clearance rate is 40.5%,protein peptides is 26.09%,polysaccharides is 90.1%;Abalone viscera hydrolysates of·OH also significantly inhibited,the IC500 of·OH is that enzymatic crude extract is 2.4 mg/mL,protein peptide is 2.1 mg/mL,polysaccharide is 7.2 mg/mL;Meanwhile,the total antioxidant capacity of abalone viscera hydrolysates was measured at a concentration of 1mg/mL,enzymatic crude extract was total antioxidant capacity was 3.55 U/mL,protein peptide is 1.91 U/mL,polysaccharides is 4.17 U/mL.In vivo antioxidant experiments results show that Three kinds of abalone viscera hydrolysates can significantly increase SOD activity in mice?P<0.01?and significantly increase in mice GSH content?P<0.05?.Compared with model group,enzymatic crude extract dose group,protein peptides middle dose group,polysaccharides middle dose group of mice SOD activity increased 59.6%,33.2%,20.57%,GSH content in mice were increased 1.5-fold,1.3-fold,1.0-fold.At the same time,it is possible to reduce the content of MDA?P<0.01?and protein carbonyl content?P<0.05?in mice,compared with the model group,enzymatic crude extract dose group,protein peptides middle dose group,polysaccharides middle dose group of MDA content in mice were reduced 48.2%,43.93%,56.2%,protein carbonyl content decreased by31.5%,55.4%,45.4%.The results show that abalone viscera hydrolysates have significant antioxidant activity in vivo.In vivo enhance immune activity experiment results show that three kinds of abalone viscera hydrolysates are able to significantly increase delayed type hypersensitivity?DTH?the extent and level of serum hemolysin in the normal mice?P<0.01?.Compared with the control group,enzymatic crude extract dose group,protein peptides middle dose group,polysaccharides middle dose group of DTH response extent in mice were increased by 75.3%,77.1%,40.3%,serum hemolysin level increased by 48.3%,27.9%,49.3%;Three kinds of abalone viscera hydrolysates can significantly improve the phagocytic index?P<0.05?in normal mice,in which the enzymatic crude extract dose group of mice phagocytic index is 6.11,the protein peptides high dose group is 6.63,polysaccharide middle dose group is 6.30,while the control group is5.71;Compared with the control group,three samples were able to significantly increase the spleen index?P<0.05?and significantly improved thymus index of normal mice?P<0.01?.Among them,enzymatic crude extract dose group,protein peptides middle dose group,polysaccharides middle dose group of the spleen index were increased by 23.9%,24.8%,21.9%;the thymus index increased by 0.9-fold,1.2-fold,1.5-fold.The results show that abalone visceral hydrolyzates have a significantly enhanced immune activity.
