In this study,the fruits of wild Rose rugose Thunb were used as experimental m-aterials,At first the content and the changing tendency of Lutein,Zeaxanthin,?-carotene,Lycopene in the flesh and the peel of strain-1 and strain-3 of wild Rose rugose Thunb were measured by HPLC.And then,the extraction process,the purification process,antioxidant properties and stability properties of carotenoids of wild Rose rugose Thunb were studied.The purpose was to understand the quality characteristics of carotenoids of wild Rose rugose Thunb,which provided reference for the product development of wild Rose rugose Thunb and selection of natural pigments.The results are as follows1.There are Lutein,Zeaxanthin,?-carotene,Lycopene in the flesh and the peel of strain-1 and strain-3 and the content of the four types of carotenoids gradually increased with the maturity of the fruits;the content of strain-1 was significantly higher than that of strain-3.For strain-1,four types of carotenoids mainly concentrated in the peel,and it is a good carotenoids source.2.The response surface methodology was used to optimize the ultrasonic-assisted extraction of carotenoids.The optimization parameters were:solid to liquid ratio was 1:135 g/mL,ultrasonic time was 98 min,ultrasonic power was 350 W,ultrasonic temperature was 55?,and the yield of carotenoids under this process parameter was 74.30 me/100g.3.The optimum technological conditions for the purification of carotenoids of wild Rose rugose Thunb with macroreticular resin were as follows:AB-8 resin was selected,the adsorption time was 2 h,the sample concentration was 0.639 mg/mL,the eluent was hexane,the sample volume was 140 mL,the eluent volume was 440 mL,and the purity increased from 0.28%to 13.93%,and increased by 49.75 times;The crude extraction and the purification contain similar carotenoid composition,Lutein,Zeaxanthin,?-carotene and Lycopene can be detected.The content of four kinds of carotenoids in the purification is higher than that in the crude extraction,and the order of content increase multiples is:?-carotene>Lycopene>Lutein>Zeaxanthin.4.The crude extraction and purification of carotenoids of wild Rose rugose Thunb had strong scavenging ability to DPPH-free radicals and ABTS.+free radicals,and showed a significant concentration-dependent trend.The IC50 value scavenging DPPH'free radical with crude extraction was 6.51 ug/mL,lower than Vc,and the ability scavenging DPPH free radicals was higher than Vc;The IC50 value scavenging DPPH*free radical with purification was 0.34 mg/mL,higher than Vc,and the ability scavenging DPPH*free radicals was lower than Vc;The IC50 value of the purification was 52.23 times higher than that of the crude extraction;The IC50 value scavenging ABTS+free radical with crude extraction and purification was 5.01 ug/mL,35.58 ug/mL,respectively,lower than Vc,and the ability scavenging ABTS+free radicals was higher than Vc;The IC50 value of the purification was 7.10 times higher than that of the crude extraction.5.With the increase of temperature,the survival rate of carotenoids in crude extraction and purification declined,after storage for 96 h,the survival rate of carotenoids in crude extraction and purification was the highest at-20?,and the survival rate were 93.98%and 97.37%,respectively.Under dark conditions,the survival rate of carotenoids in crude extraction and purification was the highest.After storage for 8 h,the survival rate were 80.16%and 97.31%,respectively.After storage for 24 h at low temperature,the survival rate of carotenoids in crude extraction and purification with 0.1 mol/L Na+?K+?Ca2+?Mg2+and Al3+were higher than 80%,while the survival rate of carotenoids in crude extraction and purification with 0.1 mol/L Fe2+and Fe3+were only about 50%,Fe2+and Fe3+had a great influence on the stability of carotenoids.The stability of carotenoids in the purification was higher than that in the crude extraction.
