Objective: The fresh use of Chinese medicine was a major feature of the clinical application of traditional Chinese medicine.There were fresh Chinese herbal medicines in some TCM formulations.But in the practical application,it was basically replaced by dry goods.For this phenomenon,this topic firstly studied the quality standard of Leonuri Herba recens,then formulated quality standard of Leonuri Herba recens to regulate the fresh Leonurus production,use and for formulating prescriptions containing Leonuri Herba recens formulation research to provide scientific basis.For a more comprehensive evaluation of the Leonuri Herba recens quality,the topic also studied the fingerprint and formulated the fingerprint of Leonuri Herba recens.Traditional Chinese medicine should focus on fresh preservation.This topic determined the best preservation technology of Leonuri Herba recens through the study of Leonuri Herba recens preservation.Methods: First of all,the topic made a study on microscopic identification,TLC,moisture,extract and content of stachydrine hydrochloride.The method of microscopic identification was paraffin section.The method of TLC was TLC.The method of moisture content was drying method.The method for the determination of extracts content was water soluble hot dip method.The method for the determination of stachydrine hydrochloride content was HPLC-ELSD.Ten batch different source of Leonuri Herba recens fingerprint was studied by using the method of HPLC-ELSD.The data were analyzed by similarity evaluation system for chromatographic fingerprint of traditional Chinese Medicine.In preservation research of Leonuri Herba recens,we used advanced scientific technology and combined with the preservation of fruits and vegetables to conducted a study on vacuum cold storage,freeze storage,controlled atmosphere storage and preservative agent storage.The fresh keeping effect was judged by the sensory changes,the weight loss rate,the corruption rate,and the content of stachydrine hydrochloride.The best technology of freezing and fresh keeping was selected by orthogonal experiment.And using the quality standard detected the Leonuri Herba recens of freeze storage.Results: Leonuri Herba recens moisture content was not less than 70%,the extract was not less than 6.0%,the content of stachydrine hydrochloride was not less than 0.20%.The topic had established the HPLC fingerprint of Leonuri Herba recens based on eight common peaks.10 batches of Leonuri Herba recens similarity were above 0.90.The method meet the technical requirement.Controlled atmosphere storage and preservative agent storage could keep good with Leonuri Herba recens in a short period of time.Vacuum cold storage had a better effect in 30 days,freeze storage had a better effect in 180 days.The best technology of freezing storage was pre-cooling 0.5h under the condition of-30 degree Celsius,and thevacuum packaging was stored under the condition of-18 degree Celsius.In the storage conditions,the inspection were in line with the quality of the proposed standard.Conclusion: The proposed quality standard method and established HPLC-ELSD fingerprint method were simple and feasible,could be better applied to the quality control of Leonuri Herba recens.Preservation technology research provided a method for the use of Leonuri Herba recens which were not affected by season and location,and also provided the scientific basis and laid a good foundation for the industrialization work.
