Pearl powder is a kind of beauty and beauty product that can delay aging and has wide application prospect.Because it can promote metabolism,enhance immunity and delay skin aging,the research work of pearl powder in recent years mainly focuses on the component analysis,the treatment of diseases and anti-fatigue and anti-aging.However,due to the complex separation and purification conditions of nanometer pearl powder,the difficulty of water-soluble protein extraction is often ignored.The results showed that nanometer pearl powder water-soluble components had strong antioxidant and whitening activities.Therefore,this paper studied the extraction and purification methods of soluble components in nanometer pearl powder by dynamic axial preparative chromatography and preparative high performance liquid chromatography with macroporous adsorption resin separation process.The main research contents and results are as follows:(1)The method of separating macroporous adsorption resin of nanometer pearl powder water-soluble components was established.Firstly,the water-soluble components was extracted from the nano pearl powder by water extraction method,and the main component protein in the aqueous extract was purified by macroporous resin adsorption method.The optimal conditions for extracting pearl protein were selected by comprehensive analytical efficiency,production conditions and economy.The adsorption resin was AB-8,the adsorption time was 4 h,the adsorption rate was 96.46%,and the desorption rate was 90.53%.Then,after separating and purifying the pearl components by the macroporous adsorption resin,the molecular weight of the pearl protein is determined by HPLC,and the molecular weight of the purified product is all less than the general molecular weight of the pearl whitening active peptide.The above results indicate that the macroporous adsorption resin can effectively separate and purify the water-soluble protein of nano pearl powder.(2)The method of whitening and antioxidant activity of water-soluble components of nano pearl powder was established.The whitening activity of the water-soluble extract of nano pearl powder under different extraction conditions was tested by single factor experiment.The three factors of liquid-to-liquid ratio,water extraction time and water extraction temperature were selected to optimize the water-soluble components of water-removed nano pearl powder.The extraction conditions were:20%solid-liquid ratio,2 hours water extraction time,and 80?water extraction temperature.The water-soluble components of nano pearl powder extracted at this time has a strong scavenging effect on hydroxyl radicals,and has strong inhibitory effect on tyrosinase,and the scavenging effect is more remarkable after purification.It shows that it is a good natural antioxidant and tyrosinase inhibitor,which can be added as a bioactive ingredient to whitening cosmetics,which has a very broad application prospect and market value(3)The method of efficiently producing proteins pearl powder of the active ingredient was established.The pearl soluble components purified by macroporous resin was used as the research object,and guided by tyrosinase inhibitory activity(ie,whitening activity),fractional separation by dynamic axial preparative chromatography,preparative high performance liquid chromatography purification,etc.Process,efficient preparation of active components in pearl powder soluble components.After separation of the pearl powder soluble components by dynamic axial chromatography,a total of 4 components were obtained,of which component 3 had the strongest whitening activity(the tyrosinase inhibition rate was>70%).The component 3 samples were purified by preparative high performance liquid chromatography to obtain a pearl active ingredient(content>90%).The high-resolution mass spectrometry and nuclear magnetic resonance spectroscopy indicated that the active component was C11H12N2O2.Peptide compounds.The results show that the preparative high performance liquid chromatography can efficiently prepare the active components in the pearl powder protein.
