Objective:The study was to determine the content of ferulic acid in plant foods based on the optimization of extraction and the establishment of a UPLC method on free ferulic acid and to estimated daily dietary intake of free ferulic acid,bound ferulic acid and total ferulic acid.Methods:The optimize extraction conditions of free ferulic acid were investigated by orthogonal design.A UPLC method was used for analysis of free ferulic acid and total ferulic acid content in 170 plant foods.Daily dietary intake of ferulic acid was estimated according to the content in plant foods and their daily dietary data derived from the summary repot on Chinese National Nutrition and Health Survey in 2010-2013.Results:1.Optimization of extraction on free ferulic acidThe optimum extraction conditions were as follows: 60-fold amount of 50% methanol(m:V,g/mL)and with ultrasonic extraction for 80 min at 50?.2.Free ferulic acid,bound ferulic acid and total ferulic acid content in plant foodsThe free and total ferulic acid content in 170 plant foods,including cereal grains(13 species),tubers(4 species),beans(7 species),vegetables(77 species),fruit(58 species),nut seeds(11 species),was measured by UPLC.The results showed that the ferulic acid content of different plant species varied greatly,while the difference in ferulic acid content was relatively small when the plant types were the same.The bound ferulic acid content and total ferulic acid content in the cereal grains were higher than that in other kinds of plant food.The ferulic acid content of whole grains is much higher than that of refined grains.Ginger has the highest content of ferulic acid in all tested foods,although there is less ferulic acid in vegetables and fruits.3.Estimation of dietary intake of ferulic acidAccording to summary repot on Chinese National Nutrition and Health Survey in 2010-2013 and survey data on dietary intake of various foods in Xinhua District,Shijiazhuang City,the daily dietary intake of ferulic acid was estimated preliminary.The daily dietary intake of ferulic acid in urban and rural residents in China is 54.70 mg/standard-person(sp)/day,and the daily dietary intake of ferulic acid in Shijiazhuang residents is 64.82 mg/standard-person(sp)/day.Cereal grains are the main dietary source of ferulic acid.Conclusions:Cereal grains are the most abundant food type with ferulic acid content and are the main contributors to daily dietary intake of ferulic acid.It is recommended that the intake of whole grains can obtain higher levels of ferulic acid.
