Aqueous two-phase extraction is a new kind of extraction method.In this study,five kinds of composing substances for aqueous two-phase system,1-methylimidazole,hexamethylphosphorictriamide,1-methylpiperidine,diethylene glycol monobutyl ether and trimethylhexamethylenediamine,were found in our lab for the first time.They all can form aqueous two-phase systems with salt like?NH4?2SO4,Na2CO3,K2CO3,K2HPO4 and KH2PO4.The cloud point were measured and phase diagrams of these aqueous two-phase systems were drawn.The flavone was extracted from Sophora japonica L.by using1-methylimidazole-?NH4?2SO4-H2O aqueous two-phase system.Through single factor and orthogonal experiments,the extraction condition was optimized.The flavone was extracted in a yield of 24.93%and a purity of76.77%,from the system with the mass fraction of 1-methylimidazole and?NH4?2SO4 34 and 17%,respectively,and the mass of Sophora japonica L.and the total aqueous two-phase system 0.9g and 20g,repectively,at a temperature of 30?and pH of 3,for 30 minutes.High performance liquid chromatography was used to detect flavone,taking rutin as an external reference,on the conditions listed as following:an octadecylsilyl column at a performan temperature of 30?,a mobile phase composed of methyl alcohol and 1%aqueous acetic acid solution in a ratio of 45:55?v/v?at a flow velocity of 1mL/min.,an ultraviolet detector with?at 257nm,and an injection volume of 20?L.Through the experiment,two different kinds of ATPS substance were selected for extracting flavone from Sophora japonica L.,respectively;and the possible mechanisms were proposed.It was predicted that this novel method could be applied in wider area in purification field than the case demonstrated in this study.
