Object: To use separate decoction and combined decoction to make Relieving Asthma and Promoting Qi Grain,compare its fingerprint chromatographic and material basis of different preparation technologies to determine the best preparation technology for it and establish its quality standard.Method: Separately make Rhododendron Dauricum grain and Folium Epimedii grain by using the separate decoction to make Relieving Asthma and Promoting Qi grain;to make Relieving Asthma and Promoting Qi Grain by separate decoction and combined decoction;to use IBM SPSS Statistics(version 20.0)to make cluster analysis on the 15 batches of Rhododendron Dauricum herbs and 15 batches of Folium Epimedii herbs;to use similarity evaluation for fingerprint of TCM(version 2012)to respectively carry out similarity evaluation on the fingerprint chromatographic of the 4 grains mentioned above;to use thin-layer chromatographic method to identify Rhododendron Dauricum herbs and Folium Epimedii herbs as well as the 4 grains above;to use HPLC method to establish fingerprint chromatographic for the 4 grains and determine the contents of the known components;compare and analyze the differences of the fingerprint chromatographic and the contents of the known components between separation decoction and combined decoction to make Relieving Asthma and Promoting Qi Grain;to determine the best preparation technology of Relieving Asthma and Promoting Qi Grain,and establish quality standards for it.Results: The 4 grains above have been made;the cluster analysis results show that 15 batches of Rhododendron Dauricum herbs and 15 batches of Folium Epimedii herbs have been divided into 4 kinds;the similarity evaluation results show the similarity of the fingerprint chromatographic of 15 batches of Rhododendron Dauricum herbs is between 0.986~0.999?the similarity of the fingerprint chromatographic of 15 batches of Folium Epimedii herbs is between 0.987~0.998?the similarity of Relieving Asthma and Promoting Qi Grain by combined decoction is between 0.993~0.999?the similarit y of Relieving Asthma and Promoting Qi Grain by separate decoction is between 0.994~0.998,all the aboves are more than 0.9,having a good similarity;The thin-layer id entification results show that the 15 batches of Rhododendron Dauricum herbs and th eir grains both contain farreol?15 batches of Folium Epimedii herbs all contain icarii n?15 batches of Folium Epimedii grains all contain icariin and icarisid I?Relieving Asthma and Promoting Qi Grain prepared in two different ways all contains farreol,?i cariin and icarisid I;the established fingerprint of Rhododendron Dauricum has 10 m utual fingerprint peaks,the known farreol has been found from it.In Rhododendron Dauricum decoction the contents of farreol are between 0.011%~0.016%,from Rhodod endron Dauricum herbs to Rhododendron Dauricum decoction the transferring rate of farreol is between 8.39%~18.29%,in Rhododendron Dauricum grains the contents of farreol is between 0.010%~0.014%,from Rhododendron Dauricum decoction to Rhododendron Dauricum grains,the transferring rate of farreol is between 85.71%~92.86%?the established fingerprint of Folium Epimedii has 8 mutual fingerprint peaks,the known icariin and icarisid I have been found from it.In Folium Epimedii decoction the contents of icariin and icarisid I are between 0.253%~0.347% and 0.023%~0.038%,from Folium Epimedii herbs to Folium Epimedii decoction the transferring rate of icariin and icarisid I are between 50.18%~56.39% and 6.00%~11.53%,in Folium Epimedii grains the contents of icariin and icarisid I are between 0.226%~0.299% and 0.020%~0.033%,from Folium Epimedii decoction to Folium Epimedii grains,the transferring rate of icariin and icarisid I are between 82.44%~92.86% and 82.61%~92.31%?the established fingerprint of Relieving Asthma and Promoting Qi Grain by combined decoction has 12 mutual fingerprint peaks,the known farreol ? icariin and icarisid I have been found from it.In Relieving Asthma and Promoting Qi Grain by combined decoction the contents of farreol?icariin and icarisid I are between 0.012%~0.023% ? 0.259%~0.343% and 0.018%~0.029%,from Folium Epimedii herbs and Rhododendron Dauricum herbs to Relieving Asthma and Promoting Qi Grain by combined decoction the transferring rate of farreol ? icariin and icarisid I are between13.19%~18.70%?51.39%~55.86% and 4.50%~8.36%,in Relieving Asthma and Promoting Qi decoction by combined decoction the contents of farreol?icariin and icarisid I are between0.010%~0.021% ? 0.226%~0.290% and 0.013%~0.024%,from Relieving Asthma and Promoting Qi decoction to Relieving Asthma and Promoting Qi Grain by combined decoction,the transferring rate of farreol?icariin and icarisid I are between 83.33%~95.00%?83.38%~90.56% and 72.22%~86.36%;the established fingerprint of Relieving Asthma and Promoting Qi Grain by separate decoction has 12 mutual fingerprint peaks,the known farreol?icariin and icarisid I have been found from it.In Relieving Asthma and Promoting Qi Grain by separate grain the contents of farreol?icariin and icarisid I are between 0.009%~0.018%?0.231%~0.284% and 0.010%~0.019%,from Folium Epimedii herbs and Rhododendron Dauricum herbs to Relieving Asthma and Promoting Qi Grain by separate decoction the transferring rate of farreol ? icariin and icarisid I are between 9.89%~14.63% ?41.32%~51.56% and 2.75%~5.07%;By comparision,we know the contents of farreol and icarisid I in Relieving Asthma and Promoting Qi Grain by separate decoction are lower than the contents of farreol and icarisid I in Relieving Asthma and Promoting Qi Grain by combined decoction,the contents of icariin show a slight difference,by calculating the similarity of the fingerprint chromatographic prepared with the two methods shows its Mark peak's similarity is 0.993 and total fingerprint peaks' similarity is 0.853,combining with the number of their fingerprint peaks we know the variety of chemical composition of grain by combined decoction is better than that by separate decoction,it is evident that the varieties of the chemical components and the contents of the known compositions of Relieving Asthma and Promoting Qi Grain by combined decoction are both superior to Relieving Asthma and Promoting Qi Grain by separate decoction;we have determined the combined decoction method is the best preparation technology for Relieving Asthma and Promoting Qi Grain,and established the quality standards for it.Conclusion:Through contrastive analysis,Relieving Asthma and Promoting Qi Grain has 12 common peaks prepared with the two methods,the Relieving Asthma and Promoting Qi grain prepared by combined decoction is,whether in varieties of chemical compositions or in contents of known chemical compositions,superior to that by separate decoction.Therefore,the combined decoction is determined for the preparations and establish quality standards for it.Preliminary analysis shows the reasons for the difference is that Relieving Asthma and Promoting Qi Grain prepared by combined decoction is completed totally under the guidance of the theory of the traditional Chinese medicine.The two herbs affect each other in the course of extraction,while the grain prepared by separate decoction is simply combined.The effect of herbs on each other happens before taken,but not totally.Whether the difference has an effect on clinical efficacy,it needs further research.
