OBJECTIVE The topics with the Duhuojisheng soup which from " Beijiqianjinyaofang " that recorded by the Tang Sun Simiao as the sample of study,Study on the preparation process of a new traditional Chinese medicine-solid decoction which based on the traditional decoction.By comparing the extract rate?HPLC fingerprinting and the contents of gentiopicroside?paeoniflorin,and osthole of Duhuojisheng soup and its solid decoction to study the difference between the duhuojisheng soup and its solid Decoction,To investigate whether the preparation process of Duhuojisheng soup solid decoction is feasible,At the same time,it provides an experimental basis for the further research of Duhuojisheng soup solid decoction.METHODS In this paper,the experiment was divided into four parts The first part The study on the preparation process of the duhuojisheng soup solid decoction:Investigating the range of various factors that affect the preparation of duhuojisheng soup solid decoction by single factor experiment and screen its optimum process conditions by orthogonal experiment and validating the best processes to ensure the preparation process is stability and feasibility.The second part Comparative study on the extract yield between the Duhuojisheng soup and its solid decoction: Extract yield is the total material basis for the clinical application of traditional Chinese medicine compounds,Comparing the extract yields of Duhuojisheng soup and its solid decoction,we can found the difference in decoction rates of effective chemical ingredients of the Duhuojisheng soup and its solid decoction.The study on the leaching rate of the solid soup of Duhuojisheng soup solid decoction can reflect the appropriate brewing time of the solid soup.The third part Comparative Study on Fingerprint of the Duhuojisheng soup and its solid decoction by HPLC fingerprint.Comparison of HPLC fingerprint of Duhuojisheng decoction and its solid decoction,It can be compared as a whole whether there is a difference in the composition of the chemical composition.The fourth part Comparative Study on the content of Gentiopicrin ? Paeoniflorin and Osthole between the Duhuojisheng soup and its solid decoction.Determination of GPS?PF and OST in Duhuojisheng soup and its solid decoction by RP-HPLC method,By determining the content of three effective chemical components in Duhuojisheng soup and its solid soup,Further comparison of the differences between Duhuojisheng soup and its solid,Providing a certain reference for the adjustment of the preparation process of the solid soup of Duhuojisheng soup.RESULTS Through single-factor investigation,it is concluded that the factors that affect the preparation process of Duhuojisheng soup solid decoction mainly include coking temperature,decoction time and additive water amount.The optimum conditions for the preparation of the solid decoction ofDuhuojisheng soup were as follows: the cooking temperature was 110 ?,the decoction time was25 minutes,and the amount of water was 255 mL.There is not significant difference between Duhuojisheng soup and its solid decoction through comparetive study on the extract yield.(P > 0.05)A total of 27 chromatographic peaks were extracted from the HPLC fingerprints of the Duhuojisheng soup and its solid decoction,the retention time of each chemical component is close,but there are some differences in peak area,using similarity calculation software 2004 A version of the correlation between the two is 0.948.A RP-HPLC method was estabished for the simultaneous determination of GPS?PF and OST contents in Duhuojisheng soup and its solid decoction,The regression equation of gentiopicroside is Y=355205X+7184.1,r=1(n=6),Gentiopicrin shows good liner relationship in the range of 38 ?g ? 950 ?g.The regression equation of paeoniflorin is Y=41448X-13163,r=0.9998(n=6),Paeoniflorin shows good liner relationship in the range of 29.36 ?g?734 ?g.The regression equation of Osthole is Y=56672X-1490.8,r=0.9999(n=6),Osthole shows good liner relationship in the range of 1.874 ?g?46.85 ?g,The average recovery rates of the three samples were 97.87%,97.46%,and 99.57%,and the RSD were 0.9%,1.43%,and 0.67%.CONCLUSION The experimental results show that the preparation process of the duhuojisheng soup solid decoction is stable and convenient.The extract yield of the duhuojisheng soup solid decoction prepared under this condition is not significantly different from that of duhuojisheng decoction,Similar chemical composition,But there are some differences in the content of each chemical component,There is no significant difference in the content of osthole in Duhuojisheng decoction and its solid decoction,The content of gentiopicroside and paeoniflorin in duhuojisheng soup solid decoction is higher than that in duhuojisheng decoction.
