Plumula Nelumbinis is one of the traditional Chinese medicines.At present,most of the studies on Plumula Nelumbinisr are focused on alkaloids,but there are few studies on the flavonoids in the Plumula Nelumbinis.Therefore,in order to enrich the chemical constituents of Plumula Nelumbinis and lay a foundation for the further discovery of new lead drugs,this paper conducted a systematic study on the chemical constituents of flavonoids in Plumula Nelumbinis.On the one hand,the structures of the extracted and isolated compounds were identified.A total of 18 compounds were identified,among which 6 compounds were isolated from Nelumbo for the first time,on the other hand,by the HPLC to establish the fingerprint of flavonoids in Plumula Nelumbinis,confirmed the main composition of flavonoids in Plumula Nelumbinis with Shaftoside and Isoshaftoside,determination of the content of them in Plumula Nelumbinis,at the same time,also adopted the DPPH free radical scavenging assay for separation of Plumula Nelumbinis flavonoids ingredients for the study of antioxidant activity.First of all,this topic in Hunan Xiangtan Plumula Nelumbinis as the research object,using 80%?v/v?of ethanol as a solvent for extraction,using D001 cation exchange resin column chromatography and type AB-8 macroporous resin column chromatography and other methods to get lotus plumule flavonoids extracts,finally by silica gel column chromatography,Sephadex column chromatography of LH-20,open ODS column chromatography,preparation of high performance liquid chromatography and other methods to extract total flavonoids lotus plumule purification,with the physical and chemical properties as well as the modern spectroscopy method for the structural identification of compounds were isolated from the structure,Finally,the structures of 18 compounds were determined as follows:Shaftoside?1?,Isoshaftoside?2?,Vitexin?3?,Isovitexin?4?,Rutin?5?,Hypericin?6?,Apigenin-6,8-glucopyranoside?7?,Icariside D2?8?,N-palmitoylethanolamine?PEA,9?,N-oleoylethanolamine?10?,N-linoleoylethanolamine?11?,Dibutylphthalate?12?,?-Daucosterol?13?,?-Sitosterol?14?,?-Sitosterol palmitate?15?,3,4-dihydroxybenzaldehyde?16?,2-?3,4-Dihydroxyphenyl?-1,3-benzodioxole-5-carboxaldehyde?17?,Stigmastane-3?-alcohol-6-one?18?.Compounds 8,9,10,11,16 and17 were isolated from Nelumbo for the first time.Secondly,it was found that Shaftoside and Isoshaftoside were the main components of flavonoids in Plumula Nelumbinis,and the contents of both were determined by HPLC.The results showed that the contents of Shaftoside and Isoshaftoside were 1.43%and 2.04%respectively.Finally,this article adopted the DPPH free radical scavenging assay of flavonoids were isolated from 1?7 studied the antioxidant activity,the experimental results show that the compounds 5 and 6 have good DPPH radical scavenging ability,and,when the concentration in the 120?mol·L-1,reached between them and the effect of ascorbic acid is,from the structure and activity relationship of compounds,which may be related to flavonoids in the amount of phenolic hydroxyl in B ring the parent nucleus and location.
