Probiotics are a group of active microorganisms that are mainly colonized in the intestinal tract, skin, and other parts of the host and play an important role in regulating the micro-ecological balance of the host's related systems. With the gradual deepening of people's research on probiotics, how to further play the role of probiotics in regulating the intestinal tract on the premise of maintaining or increasing the number of probiotics is becoming a hot topic. At present, there have been many reports that traditional Chinese medicine, especially some medicine and food homology and supplementary Chinese medicine, can effectively promote the growth of probiotics in the human body and can inhibit the reproduction of harmful bacteria in the body.At the same time, after the Chinese medicine enters the body and is broken down,it can also directly antagonize the pathogenic bacteria and enhance the body's resistance. In summary,if some Chinese medicines are used in combination with probiotic products to synergize, they can exert the therapeutic properties of traditional Chinese medicines. This thesis studies the influence of traditional Chinese medicine on the growth of probiotics in vitro, and optimizes the design of the composition of traditional Chinese medicine-probiotic composite micro-ecological preparation. Then the specific experimental content and results are as follows:The effect of Chinese medicine on the growth of probiotics. In this experiment, the effects of various traditional Chinese medicines(including tonic medicines, blood remedies, etc.)on the growth of Lactobacillus helveticus,Lactobacillus casei and Lactobacillus paracasei in vitro were studied. Our research shows that some of the supplementary Chinese medicines selected in this experiment can significantly promote the in vitro growth of Lactobacillus helveticus and Lactobacillus paracasei, and it has been confirmed by flow cytometry that they will not affect the proportion of probiotics.Polygala and green tea showed a slight inhibition of Lactobacillus helveticus and Lactobacillus paracasei;Chinese medicines such as Morinda officinalis and leek seeds had a significant growth promoting effect on probiotics. For Lactobacillus casei,a few supplementary Chinese medicines can significantly affect the growth of probiotics, and most of the remaining Chinese medicines have basically no effect on the growth of probiotics. These results are consistent with previous research conclusions. The joint use of some Chinese medicines and probiotics not only has a positive effect on the growth of probiotics, but also can use probiotics to biotransform Chinese medicines and increase the utilization of Chinese medicines. In order to maximize the synergy between Chinese medicine and probiotics, and lay the foundation for the application of Chinese medicine and probiotics in actual production,we designed and preliminary optimized the Chinese medicine-probiotic composite micro-ecological preparation.Based on the better Chinese medicine-probiotic combination screened in the previous step,we designed the Chinese medicine-probiotic compound microecological preparation formula, and aimed at 10 factors that affect the growth of the bacteria(selected Chinese medicine and peptone, glucose, yeast powder)And other medium components)were optimized using response surface method. The results of verification experiments showed that after optimization, the number of viable bacteria increased by about 3.2 times compared with the normal cultured probiotics, which can further exert the combination of traditional Chinese medicine and probiotics in regulating intestinal Functional role.Using the interaction between Chinese medicine and probiotics can help further development of probiotic health care.In this experiment,we explored the effects of different kinds of traditional Chinese medicine on the growth and activity of probiotics. It is confirmed that some Chinese herbal medicines can indeed promote the growth of probiotics, and do not affect the activity of probiotics. After the optimized design of the traditional Chinese medicine-probiotic compound micro-ecological preparation, the number of viable bacteria was improved. At present, there is not much research on the interaction between probiotics and Chinese medicine. We hope that through our research we can lay a theoretical foundation for the joint use of Chinese medicine and probiotics, and lay the foundation for the further application of the combination of probiotics and Chinese medicine in production and life.
