Okra(Abelmoschus esculentus L.)is a kind of long history agriculture crop with high value of nutrient and it can be used for food,medicine and garden ornament.It is expected to have high value of exploitation and application.It is cultivated throughout the world,especially in the tropics and subtropics.China,as a large agricultural nation,has a long history of cultivating Okra.There is over 100-year history of planting in Pingxiang,Jiangxi Province.The tender fruit of Okra has abundant mineral,protein,free amino acid,lipid,vitamin,flavonoid and mucilage like polysaccharide,pectin.Pectin is a family of complex polysaccharides,which are found in high amounts in plant primary wall.It can be used in health care effect.In order to determine pectin content in Okra,10 accession and diverse tissues from QK-6 were detected.The results showed that significant discrepancy(tender siliques as reference)was found among the 10 accession and different parts,and QK-4 had the highest level of pectin,reaching 24.05%.While young siliques from QK-6 was the richest in pectin,20.73%,higher than that in leaves,seeds and roots etc.Flavonoid can not only be used as a kind of medicine for preventing cardiovascular and cerebrovascular disease,but also have the flowing physiological activities like anticancer,anti-lipid peroxidation,anti-ading,ect.In order to determine flavonoid content in Okra,11 accession and different tissues from PK-5 were detected.The results showed that among the 11 accession,CS-1 had the highest level of flavonoid,reaching 2.71%.Additionally,caffeine could not be detected in all accessions.So far,the identification and selection for breeding have been large decided by morphological markers among the Okras in land.Due to the limited number of molecular studies focused on okras,the method of morphological markers was used to identify and select it.Though it can reflect the evolutionary relationship of plants directly,this method can not reflect the inner genetic relationship of plants.Therefore,we used the method combined morphological markers with ISSR(Inter Simple Sequence Repeat)markers to analysis the genetic diversity of 48 Okras.The 22 primers were picked for ISSR-PCR,and a total of 154 fragments were amplified with an overall average polymorphism of 54.55%.We used the 154 markers to construct the dendrogram based on the unweighted pair group method with arithmetic means(UPGMA).The results showed a high level of genetic diversity between individuals.Through the cluster analysis with results from ISSR markers,the 48 Okras can be divided into four clusters at Dice's coefficient of 0.19.This study will provide an approach to the classification and the superior seed selection of this specie.
