Okra(Abelmoschus esculentus L.)is an annual herb of Malvaceae.Okra is a kind of food,medicine,either type of flower plants.Especially in recent years,along with okra's health benefits and nutritional value are found,its planting area are growing year by year.Okra's breeding varieties are also imminent.Study of okra's genetic diversity and classification is conducive to okra germplasm's collecting,preservation,identification,innovation and utilization.Meanwhile,the extraction of active ingredient and determination of content about okra's flower is also important for the development of functional products of okra.This paper collected 94 okra germplasm which include foreign and domestic resources.By using ISSR and SRAP molecular marker to evaluate the genetic diversity and phylogenetic relationships of the okra germplasm.While,we determined four kinds of flavonoids in part of the okras.The contents and results of this dissertation mainly about:(1)Study the morphology characters of 94 okra material,at 7.36 point of in Euclidean distance,94 okra resource group can be divided into 6 categories.11 varieties of late okra are divided into a class of poly,red okra varieties clustered together.Yellow-green species clustered together,wild relatives separate cluster category.(2)Using two methods which contain ISSR and SRAP molecular markers to doing 77 okra materials research at the.molecular level.Using NTSYSPc-2.1 software to count 77 okra germplasm's ISSR and SRAP markers and using the unweighted arithmetic average number of clusters(UPGMA)method produced molecules clustering tree.When the ISSR genetic similarity coefficient was 0.81,the okra material were divided into 4 clusters,okra material were divided into 5clusters at the 0.83 point by using SRAP molecular markers.(3)Conbine ISSR molecular markers with SRAP molecular markers and doing clustering analyze.Similarity values among the 77 okra genotypes ranged from 0.66 to 0.97.at the genetic similarity coefficient of 0.83,all the materials can be divided into 6 clusters.Compare integrated clustering with a single cluster of ISSR and SRAP,integrated clustering by using multiple molecular markers has more accurate reference value.(4)Comparing methanol ultrasound methods with methanol reflux extraction methods,the results showed that ultrasonic extraction method has more advantiges over reflux extraction methods.On the foundation of single factor test,using L9(34)orthogonal test the optimum technological of okra flower extraction.The results showed that when the extracting time is 30min,extracting concentration is 70%,extraction temperature is 40? extraction times is 2,the effect is the best.(5)Using HPLC method to set up the determine method of 4 flavonoids in okra,and determine the content of flavonoids in different varieties of okra flowers.lt can provide evidence for exploring the distribution of functional components of okra flowers and screening effectiveness ingredients high okra varieties.
