Magnaporthe oryzae is a catastrophic disease of rice,and chemical control is an indispensable means.It is expected to develop a highly effective natural medicine to kill rice plague due to drug residues and pathogen resistance.Early laboratory to determine the saparasitic saponins against Magnaporthe oryzae effect,but the use of larger.Reduction of efficiency is expected to meet the needs of food production Magnaporthe oryzae prevention and control.Different fraction saponins were obtained by solvent extraction with macroporous resin and silica gel chromatography.PDA transfer to obtain sapindus exogenous bacteria,the extraction of saponins were biotransformation and chemical conversion.Determination of Magnaporthe grisea Resistance by Mycelial Growth Diffusion.The preparation condition of sapogenin saponin was 70%ethanol,the ratio of material to liquid was 1:5,70?,3 times?3.5h/times?,Concentrated under reduced pressure,extracted with n-butanol 4times?4h/times?to obtain crude saponins?1?,The yield was 33.50%;D101 resin column,followed by5BV water,3BV70%ethanol elution?2?,The ethanol elution fraction was obtained and the yield was48.97%.Ethanol eluting fraction on a silica gel column,eluting sequentially with 3BV chloroform:methanol?V/V=9:1,5:1?,Collect 5:1 elution fraction?3?and spin dry,the yield was 3.60%;Again on the silica gel column,the same procedure elution,to obtain saponin fraction 5:1Sm?4?,the yield was18.61%.The above-mentioned fraction saponins inhibited the Magnaporthe oryzae fungi EC500 of159.2516,129.6176,57.6634 and 44.6407?g/mL,respectively.Three kinds of exogenous bacteria?which were identified as Penicillium,Verticillium and Carboplasma,respectively?were isolated from the unripe fruit of erosive,and the sapogenin was transformated.100?g/mL crude saponin before and after transformation of Magnaporthe oryzae fungi inhibition rate from about 30%to 18%below;by HPLC analysis of activated saponins after conversion content of zero.The contents of active saponins were increased by 0.85,0.26,1.01,0.94 and 0.89 times,respectively,with 0.1,0.5 M HCl or 0.1,0.5,1 M NaOH respectively.The antibacterial activity of0.1MHCl and 0.5MNaOH hydrolyzate was the strongest and the EC50??g/mL?decreased from134.6170 to 77.4283 and 104.9059 respectively,and the crude saponin was hydrolyzed by the two concentrations of acid and alkali.,Its active saponin content and activity close to the effect of treatment alone,The active saponins and activity of the saponin-containing saponins were almost disappeared,and then the acetylated product was hydrolyzed with 0.5MNaOH and the active saponin was restored to63%,EC50??g/ML?was 194.9845.Chemical transformation can increase the activity of sapindosporin content,the transformation mechanism is the alkali removal of saponins in the acetyl and acid hydrolysis to remove glycosidic bonds,so that other saponins into active saponins.
