China has very wide resources of edible mushrooms,of which,many kinds of nu trients are crucial to the health of human beings,such as abundant proteins,fats,polys accharides and so on.Moreover,many researchers demonstrated that polysaccharides showed strong activities of anti-aging,blood fat reduction,anti-tumor,liver-protecting,as well as accelerating biosynthesis of nucleic acids and proteins.Therefore,Pleurotu s djamor is known as a kind of rare mushroom in edible fungi and with very high expl oiting potentials,however,related research is seldom in the world.This study optimiz ed the deep fermentation conditions of Pleurotus djamor,tested their growth state,analyzed the chemical constituents in their fruit body cultured by different solutions c ontaining certain Traditional Chinese Medicines,which would provide a theoretical b asis for the cultivation and industrialization of Pleurotus djamor.Firstly,the single-factor test of liquid culture medium of Pleurotus djamor was used to design the carbon source,nitrogen source and inorganic salt for the growth of Pleurotus djamor.The optimum culture conditions were determined as follows:glucose as the carbon source,peptone as the nitrogen source,and inorganic salts as the KH2PO4.On this basis,the orthogonal test design method was used to determine the optimal combination of the growth medium of the Pleurotus djamor mycelium was glucose 50 g/L,peptone 25 g/L,potassium dihydrogen phosphate 0.5 g/L,vitamin B1 0.15 g/L.The order of their influence is peptone>glucose>VB1>KH2PO4.Secondly,compared the biological conversion rates of cultivated red oyster mushroom fruit bodies with different drug dregs:Dendrobium,Bupleurum,Rhodiola,Rhizoma,chuanxiong,etc.as raw materials for cultivation:raw material culture>Litchi shell material dregs>Scutellaria powder dregs>Rhodiola extract dregs>Dendrobium spp.dregs>Chuanxiong spores dregs.The results showed that the yield of Pleurotus djamo fruit body cultured by different dregs had a certain effect,but the difference was not significant.The bioconversion rate of Pleurotus djamor cultured by dregs was not much reduced,but the overall impact on the fruit body of Pleurotus djamor was little.The fat content and crude polysaccharide content in the fruit body of Pleurotus djamor were further compared.The results showed that there was no significant difference in the content of fat and crude polysaccharide in the fruit body of Pleurotus djamor,which was cultured in the dregs culture and in the non-drug control group.This study showed that when the dregs content was about39%,there was no significant difference in the mushroom appearance between the Pleurotus djamor and the control group.Different dregs have a certain influence on the yield of Pleurotus djamor fruit body,but they are not much different.Due to the low price of discarded dregs,it can still reduce the production cost of production enterprises and achieve the purpose of environmental protection and waste utilization.Finally,the antioxidant activity of crude polysaccharides in fruit bodies of Pleurotus djamor cultured in different dregs and Pleurotus djamor fruit bodies without dregs were compared.The results showed that the crude polysaccharides of Pleurotus djamo fruiting bodies cultured in dregs were obtained.The crude polysaccharide concentration of 3.0 mg/ml was similar to that of vitamin C when the mass concentration was 0.08 mg/ml.The crude polysaccharide of Pleurotus djamor cultivated in dregs had a certain antioxidant activity.The crude polysaccharides extracted from Pleurotus djamor cultivated in Rhodiola,Chuanxiong and Litchi hulls and the crude polysaccharides extracted from common raw materials of Pleurotus djamor were not significantly different in antioxidant activity,indicating that these three dregs cultured,the antioxidant activity of the Pleurotus djamor fruit body had no significant effect.The antioxidant activity of Pleurotus djamor cultivated with Bupleurum and Dendrobium was higher than that of common Pleurotus djamo.The antioxidant activity of Pleurotus djamor was slightly lower,but the effect was not significant.H2O2,as a classic oxidation factor,acts directly on cells to induce cell damage and apoptosis.In this paper,vitamin C is used as a positive control.The effect of the crude polysaccharide of Pleurotus djamor cultured with different dregs on the activity of HepG2 cells induced by H2O2.The result shows that,compared with the blank group,the crude polysaccharides of Pleurotus djamor cultured in ordinary raw materials could significantly alleviate the decrease of the activity of H2O2induced HepG2 cells and has the effect of antioxidation.The crude polysaccharides of Pleurotus djamor cultured with Rhodiola,Ligusticum chuanxiong and litchi shells also have antioxidant effects.Those indicated that cultivation of Pleurotus djamor with dregs is feasible.
