The cultivating technique of liquid isolate in Harbin area, nutrition composition analysis and polysaccharide abstracting technics of fruiting body of Pleurotus djamor were studied in this paper. The cultivating technique included the culture characteristics of hyphae, screening of first-class isolate culture medium, and screening of liquid isolate culture medium, production of cultivating isolate and management technique of mushroom growth. Many items of nutrition composition of Pleurotus djamor fruiting body cultivated were analysed, polysaccharide abstracting technics and polysaccharide function of it was studied. Total results are as follows:(1) Results of the culture characteristics and the screening of first-class isolate culture medium indicated that both of the two tested culture media were fit for the growth of Pleurotus djamor mycelium, hyphae grew very well, white, villiform, cotton wool shape to powder shape. The mycilium of Pleurotus djamor could produce extracellular phenol oxidase. From the point of economical view, PDA was the most suitable first-class isolate culture medium for Pleurotus djamor. The optimum temperature for the growth of hyphae was 25℃~28℃, the optimum pH is 6—7.5 kinds of liquid media were used in the culturing of second-class isolate. Through the contrasts on the hyphae mass of liquid isolate, hyphae's growth condition and days of overgrowing the nutrient fluid, concluded that the optimum liquid media for Pleurotus djamor were cornmeal-dextrose-peptone and bran-dextrose-peptone media, mycelia could overgrow the nutrient fluid in 7 days.Contrasting second-class liquid isolate as mother isolate for Pleurotus djamor to produce cultivating isolate with solid original isolate as mother isolate, the former had the merit of hyphae restoring quickly, hyphae growing fast in solid medium, hyphae growing well and hyphae growing uniformly. The general method similar as Pleurotus ostreatus was adopted to make cultivating isolate, treat hyphae growth and manage mushroom growth. For adopting the cultivating model of liquid isolate instead of solid original isolate as second-class isolate, made the cultivation circle of Pleurotus ostreatus cut short greatly, at least above 16 days.(2) The analysis result of Pleurotus ostreatus fruiting body nutrient composition indicated that the moisture content of fresh fruiting body was 90.16%, the moisture content of natural air seasoning dry sample was 6.36%, fat content was 2.63%, protein content was 24.69%, acidity was 0.1873%, total ash content was 9.43%, reducing sugar content was 0.507%, total content of amino acid was 17.6%, contained the human body essential amino acid eight kind of center seven kinds, including isoleucine, leucine, phenylalanine, lysine, threonine, amino-isovaleric acid, methionine, as well as other several kinds of non-essential amino acids. For adopting themethod of acid hydrolyzation analyze amino acid composition and content, tryptophan hadn't been detected out, also mensurated the content of VBi, VB2, VC and VE, separately was 0.032mg/100g, 0.469mg/100g, 0.20 mg/lOOg, 701.16 mg/lOOg, mensurated the contents of some mineral elements, including mass elements and trace elements. In mass elements, the content of kalium was 1292mg/100g, the content of phosphorus was 1206 mg/lOOg, the content of natrium was 338.06 mg/lOOg, the content of calcium was 151.44 mg/lOOg, and the content of magnesium was 262.25 mg/lOOg. In trace element, the content of zinc was 161 mg/lOOg, the content of iron was 10.89 mg/lOOg, the content of cuprum was 15.28 mg/lOOg, and the content of manganese was 1.46 mg/lOOg.(3) Adopting the method of hot water abstracting, abstracting technics of Pleurotus djamor polysaccharide was studied;the elementary experiment on the fuction of abstracted Pleurotus djamor fruiting body polysaccharide was also done. The result indicated that the best abstracting technics of Pleurotus djamor fruiting body polysaccharide was adding water 50 times, 90 °C, abstracting 3 hours, abstracting 3 times, in this case, the abstracting rate was highest. When used ethanol deposit polysaccharide, adopting ethanol's concentration of above 70% could achieve better effect. Adopting the method of coomassie brilliant blue G-250 mensurate the protein content in gross polysaccharide was 4.62%, adopting Sevag method eliminate the dissociative protein, the polysaccharide after eliminating protein via H2O2 decoloring, gained the Pleurotus djamor polysaccharide slightly appeared pale yellow. The test result of polysaccharide function indicated that the Pleurotus djamor polysaccharide had the functions to eliminate hydroxyl free radical and super oxygen ion free radical, had better eliminating function to super oxygen ion free radical. But the inhibiting function to Echerichia coli DH5a wasn't evident;it hadn't inhibiting function to Aspergillus niger.
