| As a new industry,the donkey industry has gradually been valued by people.Its peripheral research has gradually begun to enter various laboratories and become a new hot spot.In order to investigate the pathogenicity of transportation to donkeys and the impact of transportation on the physiological,blood,biochemical and hormonal parameters of donkeys,this study used 5-month-old donkey cockroaches as the object of study,and its weight changes after transportation.Incidence,hormone indicators,blood physiology,biochemical indicators,and stress protein expression indicators were measured and statistically analyzed.Trial Ⅰ.Investigation on the pathogenicity of donkey pupae during transport stressIn this experiment,two groups of healthy donkeys were selected from different places to be transported back to the Shandong breeding base.Isolation and rearing was followed for a month to detect the morbidity and mortality of the donkey population.The first batch of experimental animals:In May 2017,ten male donkeys from Fuxin,Liaoning province were in good health and had an average weight of 126 kg;the second batch consisted of 24 well-selected male donkeys from Chifeng City,Inner Mongolia,in December of the same year.Weight 144kg;age of two batches of donkeys averaged 6 months.After reaching the Dong’a County in Shandong province,the weight,incidence and blood physiology and biochemical indicators of these two groups of donkeys were tested continuously for one month and statistical analysis results were obtained.The following results are obtained:(1)The incidence of first donkey warts was 30%;the incidence of second donkey warts was 37.5%.The mortality rate of the first batch of donkeys was 10%,and that of the second wholesale was 0.The first batch of donkeys lost an average weight of 8.9 kg before they got off the train and the second batch of donkeys lost an average of 5.9 kg.(2)Results of blood routine test before and after donkey transport:The difference between Neu(neutrophil)transport and pre-transportation was extremely significant(p<0.01),and the difference between eosinophils before and after transport was extremely significant(p<0.01).Lym(lymphocyte)decreased significantly compared with before transport(p<0.05).(3)Statistic results of blood biochemistry before and after transport of donkey’s beak:ALB and GLU in serum of donkey were higher than those before and after transport(p<0.01);transport resulted in severe damage to donkey diaphragm muscle cells,and in muscle cells.Creatine kinase CK and AST in muscle cells escaped,which significantly increased serum CK and AST levels(p<0.01).Test Ⅱ Effect of Three Antistress Treatments on Donkey’s Resistance to Transport StressA total of 40 male and female donkeys with similar clinical examinations(about 130kg)and about 6 months of age were selected for the trial.The donkey cockroaches were randomly divided into four groups.The test group A was treated with Chinese medicine plus five-masculine and antibiotics,and the test group B was fed.probiotic biological agent preparations,experimental group C drinking branch original net,electrolytic multidimensional and antioxidant mixtures,the control group did not do any treatment.Temporarily processed for 3 days and shipped to Shandong Dong’a.The collected samples were mainly used for statistics of body weight changes of donkeys,blood physiology,biochemistry,and protein expression indicators.The following results were obtained:(1)Transport causes severe body weight loss,and has a strong and persistent effect on the growth and development of donkey’s late stage;after 12 hours of transport,the donkey’s weight loss of each group is compared with group A<group C<group B<control group,after transportation The 30-day weight change of donkeys was found in group A>group B>group C>control group,and after 30 days,the body weight of donkeys basically recovered to the level before stress.(2)In the control group,the RBC,HGB,Neu,and WBC in the blood of the donkey were all significantly increased(p<0.01),but Lym was also significantly lower(p<0.01).The trend of changes in the corresponding indicators of the test group A and the pre-transportation group was the most stable,and the change trend of the group C relative to the group B was relatively slow.(3)The biochemical markers Ca,ALB,Tp,GLU and CK were significantly higher(p<0.05)in the control group than in the post-transport period(p<0.05).The CK recovered to pre-transport 5 days after the operation.Levels,other data basically reached the same level before transportation after 3 days after transportation,and the CK change results showed the highest value on the day after transportation.After 12 hours of transportation,the indexes of the control group and the test group A were significantly different(p<0.01).The trend of biochemical indicators in the three groups of trials was slower than that of group B,and there was no significant change in group A.The trend of group C was slightly slower than that of group B,but the difference was not significant.(4)The levels of Cor hormone and HSp90 protein in the control group increased significantly at 12h post-transport,and returned to pre-transport level 3 days after retransmission.After 12 hours of transport,Cor and HSp90 expression levels in the control group were significantly higher than those in the test group A.Extremely significant(p<0.01).The change trend of the test group A before and after transportation was slower than that of the group C,followed by the group B.Through experimental analysis and comparison,the following conclusions aredrawn:1.The stress caused by transportation caused the difference of Lym in the blood routine of the donkey quail after 12 hours of transportation was extremely significant(p<0.01),Neu increased significantly after 12 hours of transportation(p<0.01);serious imbalance in the transporter was observed.In the state of donkey,the muscle tissue of the donkey was severely damaged,and the CK index in the serum was significantly increased(p<0.01)at 12h after transportation;the concentration of cortisol in the donkey was significantly increased(p<0.01)after peripheral transportation.The amount of HSp90 expression in cells was also significantly(p<0.01)higher after transport.2.The effects of three anti-stress treatment programs on donkey puppies showed that the effect of combined use of antibacterial agents and antibiotics was superior to the combined effects of adding electrolytic multidimensional,antioxidant and Zhiyuanjing,and electrolytic multidimensional antioxidants and Zhiyuan.The net anti-stress effect is better than the addition of probiotic biological factors. |
