Four experiments were conducted to determine the digestion of dietary nitrogen metabolism, the degradability of dietary component, the rumen environment, and the urea transporter-B(UT-B) relative quantity expression in gastrointestinal tract tissue of various dietary in the Inner Mongolia White Cashmere goats. Findings can be regulated by dietary UT-B expression in the gastrointestinal tract.Exp 1 The effects of various dietary on digestion and metabolism of nitrogen in the Inner Mongolia White Cashmere goats18 Inner Mongolia White Cashmere goats with the same father(initial BW=34.95±2.37kg) were blocked by initial BW into 6 group, fitted with permanent cannulas in the rumen, with supplements cracked(C) and swelling(S) corn two types dietary, that inclusion of three crude protein level,7.5%(L),10.5%(M) and 13.5%(H). The result were: nitrogen metabolism in goats were impacted by dietary type and crude protein.The nitrogen intake, degradable nitrogen, retained nitrogen and retained nitrogen/metabolic body weight were increasing with the dietary crude protein increasing. Retained nitrogen and the ratio of retained nitrogen/metabolic body weight were significantly raised, but the plasma ammonia nitrogen were markedly reduced by swelling corn dietary. We conclude that swelling corn-based dietand high crude protein could promote the deposition the dietary nitrogen in animal body.Exp 2 The study of digestibility of various dietary in the Inner Mongolia White Cashmere goatsThe animal was the same as Exp 1.In the Exp, the apparent matter, dry matter(DM), crude protein(CP), neutral detergent fiber(NDF), acid detergent fiber(ADF) of various dietary were evaluated by nylon bag.The trail used"successive run,one out"strategy,and put the nylon bag at 0,2,4,6,8,10,12 hours after morning feeding. The result are as follows, the digestibility of apparent matter,DM,CP,NDF and ADF were rising with extension of experimental time. Medium and high dietary crude protein remarkably enhanced the digestibility of apparent matter and its ruminal kinetic parameter P and C, and the digestibility of DM and its ruminal kinetic parameter B, A+B,P and C, and the digestibility of CP before 8 hour. Low and medium protein of dietary could notedly boost ruminal kinetic parameter A, A+B, and P of NDF. Nevertheless, ruminal kinetic parameter A+B were markedly ascended by low and high protein of dietary, kinetic parameter C were particularly rising with crud protein increasing. Swelling corn type dietary had higher apparent matter ruminal kinetic parameter A and P, DM, CP and ADF relative kinetic parameter P, NDF relative parameter B and P, than cracked corn type dietary, however, it could reduce the digestibility of apparent matter and NDF ruminal kinetic parameter C.Conclusion:High crude protein and swelling corn could increased the digestibility apparent matter,DM,CP(before 8 hour) and NDF, and their's ruminal kinetic parameter P,so as to promote the digestion of dietary.Exp 3 The effects of various dietary on the rumen environment in the Inner Mongolia White Cashmere goatsThe sample of ruminal chyme were collected at 0,2,4,6,8,10,12 hour after morning feeding, and measured pH,urea-N, ammonia-N(NH3-N) and volatile fatty acids(VFA). The finding was that pH and acetic acid / propionic acid were grasdually increasing,but other indictators include urea-N, NH3-N, acetic acid, propionic acid,butyric acid and total volatile fatty acids(TVFA) had downward trend in varying degress, with erprimental time postponeding.With rational crude protein enhancing and swelling corn instead cracked corn,the samples'pH, urea-N, NH3-N, acetic acid, propionic acid, butyric acid and TVFA had increased, and the ratio of acetic acid/propionic acid has decreased. However the pH was turning down when the protein were low level,and swelling corn type ration could boost the acidity of the samples. We made a result that the urea-N, NH3-N, acetic acid, propionic acid, butyric acid and TVFA of ruminal samples would been increased by high crude protein and swelling corn ration, with ratio of acetic acid/propionic acid lowering.Exp 4 The effects of various dietary on UT-B expression in the Inner Mongolia White Cashmere goats'gastrointestinal tractsThe goats were in a completely randomized selected in each group,euthanized and samples taken for Western blot analyses for UT-B,and evaluated the relative quantitative of the UT-B protein 30~34kD.The results showed that immunobloting confirmed the presence of 30~34kD UT-B bands in all 72 tissues analyzed, but there were some other protein bands in each sample with the ration changing. Comparing various samples,we found that the compound stomach were evidently higher than intestinal tract,the liver was notably more(P<0.05)than the parotid gland, significantly higher(P<0.01) than kidney.The high crude protein level of ration could impetus the expression of UT-B protein in fore-stomach,ileum, cecum,caecum,and kidney, while inhibit expression of the protein in duodenum, however,in the parotid gland, abomasum,the liver was less affected. Swelling corn ration could highten the protein expressing in abomasums,duodenum, cecum,caecum,and kidney,and depress the UT-B expression in ileum and liver.
