China,as an antibiotic producing and using big country,produces a large amount of drug residue in the production,it is easy to generate resistance genes under environmental stress,and then seriously threatens human health.Therefore,in 2008,China identified the antibiotic residue as the “National Hazardous Waste List”.Dangerous solid waste.Therefore,the company urgently needs to find a solution to the efficient and harmless disposal of antibiotic residue.Mainly around the establishment of spectinomycin residue residue detection method,to explore the impact of mixed aerobic composting of spectinomycin residue and sludge on physical and chemical parameters,biological characteristics of the heap,and finally assess the composting product maturity and safety.It is expected to be widely used in production practice.In order to accurately determine the residual spectinomycin in the residue,an ultra-high performance liquid chromatography-tandem quadrupole mass spectrometry for the determination of spectinomycin in the residue was first established.The solid phase extraction method was selected as the pretreatment method for spectinomycin residue in the residue,and the necessary solid phase extraction pretreatment and chromatographic conditions were optimized.Finally,the mixture of trichloroacetic acid and ammonium acetate was used as the spectinomycin residue.The extractant was used as the activator of the solid phase extraction column with a volume of 5ml of water and methanol,respectively.The volume of 5ml of water and methanol were used as the elution agent of the solid phase extraction column,and finally the solid phase extraction eluent.Use a volume of 5 m L of 10% ammoniated methanol.The best precursor ion of spectinomycin and corresponding secondary ions with high response values were 333.10 and 98.16,corresponding to the best detected cone voltage and collision energy were 20,36;30,18,and the mass spectrometry conditions were: Positive ion scan mode,multiple reaction monitoring,cone voltage 30 v,ion source temperature 350°C,cone gas flow 35L/hr,solvent removal flow 700L/hr,ion source capillary voltage 3.5kv.Chromatographic conditions were eluted isocratically with acetonitrile-0.1% formic acid water at a ratio of 80/20(v/v).When the addition level was 50-2000 mg/kg,the range of intra-day and inter-day recoveries of the method met the requirements.The detection and quantitation limits of the method were 0.06,0.1 mg/kg,respectively,and the linearity was good.It shows that this method can be used as a method to detect spectinomycin residue in the residue.The experiment was conducted by mixing aerobic compost with the spectinomycin residue and the sludge,and by investigating the changes of the basic physical and chemical parameters at three different bath temperatures,the degradation of spectinomycin,and the study of microbial properties such as enzyme activity,bacteria and other culturable microorganisms.The number of changes verified the feasibility of aerobic composting of urban sewage sludge mixed with spectinomycin.The results are as follows: The odor of the heap body gradually decreased from the initial pungent odor until it disappeared,and the body color changed from the initial light yellow to dark brown.In the early stages of composting,the temperature of three reactors increased significantly.The trend was to meet the variation of aerobic composting temperature;the moisture content of the three reactors decreased by about 30%;the final p H values were 7.57,7.78,and 7.96.Comply with compost maturity for p H Requirements,the final body is weak alkaline.The last p H of the body 3 is highest.The total organic carbon content continues to decline with the progress of composting.The change trend of total nitrogen content is basically the same as total nitrogen content.The body 3 descended significantly faster than the first two stacks.In the early stages of composting,the seed germination rate was almost 0,and after composting it exceeded 50%.At the same time,the content of spectinomycin in the compost is almost degraded to zero during the composting cycle,indicating that the aerobic composting method can realize the harmless treatment of the compost.In the study of the bio-biological properties of the heap body,bacteria play a major role in the heap,so the number of bacteria is the largest,and the number of actinomycetes and fungi is small.From the experimental data analysis of the enzyme activity of the compost,the activities of catalase and cellulase were relatively high,while the activity of invertase was low.The enzyme activity shows a corresponding regularity with the composting time and the bath temperature.The data in the three stacks indicated that the number of bacteria and actinomycetes in the culture medium was basically the same,both in the early stages of composting,and the fastest growing rate.The number of bacteria gradually decreased when entering the endogenous respiration stage;the fungi were more susceptible to the living environment.The impact of the decline in the number of the first with the temperature,showing a rising trend in the later period.Finally,the quality of the compost product was evaluated by three-dimensional fluorescence,nutrient element analysis and E.coli quantitative analysis in the compost.The quality of the compost product complied with national agricultural industry standards.The number of major pathogen E.coli in the compost body was also almost zero,indicating the compost product.Safety,the above experimental results prove that the aerobic composting method is feasible to deal with the harmless treatment of spectinomycin residue.
