Anemone roxburghii is a traditional Chinese herbal medicine,which has important medicinal value in the treatment of hypertension,diabetes and cancer.At present,the market demand of Anemone roxburghii is increasing year by year while the products are mixed.Therefore,the industrialization development of high-yield and high-quality in Anemone roxburghii is becoming an inevitable trend.Tissue-cultured bottle seedlings of Fujian Anemone roxburghii and Mingxi Anemone roxburghii,widely popularized,were selected as experimental materials,which were testified after consulting a large number of documents.The results showed that:(1)The indexes of Anemone roxburghii tissue-cultured bottle seedlings,like plant height,stem node number,leaf number,root number and yield,could be significantly increased with the help of greenhouse substrate cultivation,especially in the first month.After three months,the yield could be increased by more than one fold.The content of polysaccharides and flavonoids in Anemone roxburghii could be decreased at the initial stage of cultivation in greenhouse,but the content of polysaccharides and flavonoids would gradually increase with the prolongation of cultivation time;(2)The best matrix formula for Anemone roxburghii tissue-cultured seedlings was that soil:vermiculite:pine bark = 7:2:1 with a great increasing of yield and quality,particularly in fresh and dry weight,23%in Polysaccharides.Besides,the results showed another matrix formula:fine:sand = 4:1 could markedly increasing the yield.with the resh and dry weight could be increased by 2 times,but the effective ingredients were relatively low,which could be solved by prolonging cultivation time;(3)Salicylic acid(SA)could improve the yield of Anemone roxburghii.,and high concentration was the best treatment for two species of Anemone roxburghii which cultured at room temperature,while low concentration was the best treatment under high temperature stress.High concentration of SA could increase the polysaccharide content of Anemone roxburghii no matter the plants cultured at room temperature or high temperature.Low concentration of SA could promote the content of total flavonoids relatively well,while medium and high concentration of SA could increase the content of total flavonoids by increasing the activity of PAL enzyme.
