Takifugu rubripes(?)and Takifugu flavidus(?)breed hybrid offspring Jiyan-1 fugu.In terms of quality,the taste of Jiyan-1 fugu similar with Takifugu flavidus,and the market value is higher than Takifugu rubripes.In farming,the growth rate of Jiyan-1 fugu faster than Takifugu flavidus',closely with Takifugu rubripes',and its has high survival rate than the parents,generally about 90%.In terms of resistance,Jiyan-1 fugu has strong resistance to heavy metal ions,water temperature and salinity fluctuations.However,research carries out less about hybrids' biological and genetic characteristics,so this paper carry out basic research from two parts: the genetic diversity between Jiyan-1 fugu and its parents,in response to pathogens stimulate the body.Results are as follows:By the method of SSR,to analyze the genetic diversity between Takifugu rubripes,Takifugu flavidus and Jiyan-1 fugu,by screening the primer design,we can get 15 pairs of SSR primers which have specificity had polymorphism and express favorable generality at species level.By cluster analysis,Na(6.466667),Ne(3.2096),Ho(0.56)and He(0.592)are distributed between parents.Cluster analysis showed hybrids distributed between the parents,whose similar proportion and similar value with maternal are slightly higher.According to the method of perperitoner injection with Edwardsiella tarda in Jiyan-1 fugu.During the 36 h,the value of AKP,ACP,T-SOD increased(P<0.01)in liver,the value of GSH-PX and GSH increased in the same.During that time,ACP decreased,and the value of GSH-PX,T-SOD are lower than the control(P<0.05)in kidney,besides,MDA increased first then decrease.After injection with vibrio harveyi,the value of GSH-PX,T-SOD are higher than the control,and GSH was increased,to the opposite,MDA was decreased in liver.After injection,ACP,GSH-PX and T-SOD are lower than the control(P<0.05)in kidney,besides,MDA increased first then decrease.By peritoneal injection of Edwardsiella tarda and vibrio harveyi,real-time PCR was used to analyze the changes of immune-associated genes RAGs,LCK,MHC II in 72 h.In liver,RAGs which were stimulated by Edwardsiella tarda,the relative expression of them were obviously decreased,peaking at 6 h after injected.In spleen,the relative expression of RAGs were increased first then decrease,which was peaking at 6 h after injected.In kidney,the relative expression of RAGs were increased first then decrease,but they appears to rebound in the 48 h,and the maximum values of RAG1 and RAG2 m RNA appeared at 48 h and 6 h,respectively.RAGs which were stimulated by vibrio harveyi,the relative expression of them were obviously decreased in liver,and the maximum values of RAG1 and RAG2 m RNA appeared at 12 h and 72 h,respectively.In spleen,the relative expression of RAGs were increased,which was peaking at 72 h after injected by vibrio harveyi.However,RAGs which were stimulated by vibrio harveyi in kidney,the relative expression of them were decreased,and the maximum values of RAG1 and RAG2 m RNA appeared at 12 h and 6 h,respectively.Meanwhile,we verify the guessing that the expressional tendencies between RAG1 and RAG2 are basically the same.LCK which were stimulated by Edwardsiella tarda,can be expressed in liver,spleen,the relative expression of them were obviously decreased and they reached its maximum at 6 h after injected.In kidney,the relative expression of LCK was increased first then decreased,and the maximum values of its appeared at 6 h.LCK which was stimulated by vibrio harveyi in liver,spleen and kidney,the relative expression of LCK were obviously decreased,which was peaking at 72 h after injected in liver and spleen,besides,it was peaking at 6 h after injected in kidney.After infected by Edwardsiella tarda and vibrio harveyi,MHC II can be expressed in liver,spleen and kidney and the relative expression of it was obviously increased.Besides,after infected by Edwardsiella tarda,the relative expression of MHC II was peaking at 18,72 and 60 h in liver,spleen and kidney,respectively.After infected by vibrio harveyi,the relative expression of MHC II was peaking at 6,18 and 36 h in liver,spleen and kidney,respectively.
