Abstract/Summary:
In the process of soybean food processing,polyunsaturated fatty acids such as linoleic acid and linolenic acid in seeds are easily oxidized by seed lipoxygenase isozymes to produce some unpleasant smell of odors,which is a component that must be removed in food processing;β-conglycinin has a significant effect on reducing the body oil,blood lipids and blood sugar in the human body in the 7S globulin subunit.The 7S globulin content in soybean is significantly negatively correlated with the 11 S globulin content,and the oil is created by genetic polymerization.The double-deletion traits of oxidase and 11 S globulin subunits to obtain genetic material with no beany flavor and significant "three high" physiological functions,and provide specific parents for the cultivation of high value-added soybean cultivars,which is the main target of this study.In this study,the lines "HI8005","HI6934",and "Z+7S+I-",which are completely deleted by lipoxygenase(Lox-1,Lox-2,Lox-3),are used as the female parent,and the 11 S globulin subunit(I,IIa,IIb)completely deleted the high 7S globulin content of the strain "Na Na" as the male parent,respectively prepared cross combination by artificial hybridization and other gene polymerization methods,created the newly developed ISDS-PAGE modified polyacrylamide gel electrophoresis technique,the 496 F2 seed 11 S globulin subunit deletions were identified,and 11 seeds of 11 S globulin subunit deletion F2 seeds were obtained for nutrient steroidal potting.The cotyledons were cut into 20 mg,then the lipoxygenase deletion analysis was performed by ISDS-PAGE until the cotyledon was unearthed and before the true leaves were unfolded.Five parts of the F5 soybean lines with the double-deletion characteristics of the lipoxygenase and the 11 S globulin subunit,two parts of the complete deletion of the lipoxygenase,three parts of the 11 S globulin subunit deletion and one part of the lipoxygenase and the subunit I of 11 subunit deletion were finally obtained.Through the obtained 11 S globulin subunit and lipoxygenase double deletion material,lipoxygenase and 11 S globulin subunit single deletion high-generation materials to analyze and detect the main quality traits such as seed protein and the components of amino acid,oil and fatty acid,and biostatistics were obtained from the experimental data.Analysis to explore the effects of different subunit deletions on the above-mentioned major quality traits.This study draws the following results:(1)The newly developed soybean lipoxygenase deletion seedling cotyledon rapid detection method can be applied to screening of its hybrid progeny and germplasm resources in which soybean lipoxygenase is completely deleted or Lox-1,Lox-2 and Lox-3 are missing.It perfectly solves the problem that the traditional precaston cotyledon detection method cannot ensured safe seedling emergence and reproduction breeding of the target seed due to damage of the seed cotyledon of the seed;(2)The protein content of lipoxygenase and 11 S globulin subunit deletion experimental material was higher than that of the control variety "Heihe 38",which was 12.45% higher than the latter,the percentage of 7S globulin in the experimental materials of lipoxygenase and 11 S globulin subunits was increased by 150% compared with Heilongjiang Province’s main soybean variety "Heihe 38",The lysine content of the experimental materials of lipoxygenase and 11 S globulin subunits was significantly increased,which was higher than that of all control varieties,which was 10.6% higher than the control variety "Heihe 38";(3)The oil content of lipoxygenase and 11 S globulin subunit deletion experimental material was lower than that of the control variety "Heihe 38",The content of saturated aliphatic acid in lipoxygenase and 11 S globulin subunits was low,and the content of oleic acid in unsaturated aliphatic acid was higher than that of the control variety "Heihe 38",linoleic acid content and other;(4)Comparing the content of protein,oil,fatty acid and amino acid between double-deletion materials and single-deletion materials and their control varieties,the content of oil,palmitic acid,stearic acid and oleic acid among different experiment materials.The variation is large,which lays a material foundation for the creation of hybrid parents with different missing traits.Therefore,The newly developed rapid identification method of soybean lipoxygenase-deficient cotyledon at seedling stage can effectively solve the bottleneck problem of soybean quality improvement breeding that the traditional method of cotyledon detection before sowing cannot ensure the safe emergence of target seeds and reproduction of offspring due to the damage of cotyledon of tested seeds.Using the new method for screening for the fat enzyme and 11 s globulin of base pairs of missing materials in beneficial to human body such as protein,lysine,oleic acid functional nutrient content,compared with the control varieties,with a significant increase in the trend,a downward trend in fat for food processing soybean varieties breeding special high added value,providing theoretical basis and the specific parents.
