| Trauma is a common case in veterinary clinic,and diabetes is one of the important factors affecting wound healing.Elevated blood glucose will lead to abnormal metabolism of cells around the wound and stenosis and occlusion of the angiogenesis,as well as a decrease in autoimmunity.Aloe vera gel has antibacterial effects,immune regulation,analgesia and promotion of granulation tissue formation,which can effectively promote wound healing.The purpose of this study is to explore the role of aloe gel in the process of skin wound healing in diabetic dogs,and to preliminarily explore on which indicators of aloe vera gel affect the rate of wound healing,so as to provide new ideas for the future clinical treatment of diabetic animal wound healing.Six dogs were fed adaptively for 2 weeks.The diabetic model was established before the traumatic model was established.Sixteen incisions were made on the dog back.From left to right,the wounds were divided into blank control group(K group),vaseline gro up(F group),aloe vera gel group(L group)and rubber raw muscle paste group(X group).Drugs were given every day and weighed once a week;recorded the healing on the 0th,3rd,7th,14 th,21st and 28 th day after operation;Image J calculated the wound healing rate,wound complete healing time and scar area on 3,7,14 and 21 d.Skin tissue samples were collected on day 7,14,21 and 28 d for HE staining and Masson staining.The levels of IL-1β,IL-6,TNF-α and TGF-β in tissue homogenate were determined by ELISA kit.Extracted tissue RNA and RT-PCR was used to detect the related factors of wound healing,such as VEGF and PDGF.Experimental results: The diabetes model and the trauma model were s uccessfully established.After 3 weeks,the weight of dogs showed a downward trend.Compared with group K,the tissue and the epithelium were generated faster in group L and X,and the scar time w as reduced.And the wound healing rate increased with time.The levels of IL-1β,IL-6,TNF-α and TGF-β in skin homogenates decreased gradually.Compared with group K,the difference of IL-1β was significant in group L(P<0.05)and extremely significant in group X(P<0.01)at 7d.At 14 d,there was a significant difference between the group L(P<0.05)and the X(P<0.01).At 21 d,only group L had significant difference(P<0.05).At 7d,the difference between the group L and the X was extremely significant(P<0.01).At 14 d,the difference between group L and X was significant(P<0.05).The TNF-α was significantly different between the group L and the X at 7 days(P<0.05).At 14 d,the difference between the group L and the X was extremely significant(P<0.01).The difference of TGF-β between group L and X was significant at 7 d(P<0.05).There was significant difference in group L at 14 d(P<0.05),and the difference between group X was extremely significant(P<0.01).The results of HE staining showed that there were a large number inflammatory cell s infiltration,a small amount of granulation tissue coverage and vascular tissue regeneration in the four groups of wounds at 7 d,but the density of inflammatory cells in group L and X was lower than that in group K and F.At 14 d,a small amount of epithelial cells covered the wound surface of the f our groups.Compared with the group K and the F,the group L and the X had fewer inflammatory cells but more neovascularization,accompanied by a small amount of fibroblasts and collagen fibrogenesis.At 21 d,a large number of mature granulation tissues wer e observed in the four groups.There were still a small amount of inflammatory cells infiltrated in the group K and F.The marginal tissues of group L and group X were completely epithelialized,with more fibroblasts and collagen fibers.At 28 d,mature granulation tissue was found in all the four groups,and epidermal and sebaceous gland regeneration was found in group L and X,but not in group K and F.Masson staining showed that the four groups of wound collagen fibers increased with the treatment time.At 7 and 14 d,the collagen fibers in group L and X were more than those in group K and F.At 14 d,collagen fibers of group L and X were evenly distributed and relatively orderly arranged,while collagen fibers of group K and F were irregularly arranged and curled into a group.At 21 and 28 d,collagenous fibers of group K and F were still in a disordered arrangement,while the staining characteristics and arrangement of collagenous fibers of group L and X were closer to normal skin tissue.The expression levels of PDGF and VEGF genes showed a downward trend as a whole,but the group showed an upward trend.Compared with group K,PDGF gene expression level was significantly different in group L(P<0.05)and significantly different in group X(P<0.01)at 7 d.At 14 d,there was a significant difference between group L and X(P<0.05).At 21 d,only group X showed significant difference(P<0.05).The expression level of VEGF gene was significantly different in the group L(P<0.05)and the group X(P<0.01)compared with the group K at 7 d.At 14 d,there was a significant difference between group L and X(P<0.05).The experimental results showed that aloe gel could accelerate the wound healing rate,shorten the wound healing time,reduce the scar area after healing,inh ibit the secretion of inflammatory factors such as il-1β,il-6,TNF-α and TGF-β,stimulate the expression of wound healing related cytokines such as VEGF and PDGF in diabetic dogs’ wound healing process,so as to accelerate the wound healing. |
