Objective To further strengthen the standardization of Peucedanum praeruptorum Dunn,improve the germination rate of Peucedanum praeruptorum Dunn seeds and establish the quality grading standard of them,the operation specifications of artificial cultivation of Peucedanum praeruptorum Dunn seeds were studied,and the planting operation rules were formulated to improve the yield and quality of Peucedanum praeruptorum Dunn and to provide a scientific solution to the current situation of resource shortage.Methods 1.Seed quality grading standards were established through the determination of morphological character,seed purity,water content,1000-seed weight,viability,germination rate,etc..2.By studying the influence of temperature,light,substrate,and different exogenous growth regulators on the germination of Peucedanum praeruptorum Dunn seeds,the main factors affecting the germination of them were screened to find the best method for improving the germination of them for agricultural production.3.According to the environmental characteristics of the provenance base,the environmental characteristics of the artificial planting base of Peucedanum praeruptorum Dunn were investigated,and the air,water quality and soil quality of the planting base were studied,so as to improve the artificial planting base of Peucedanum praeruptorum Dunn;according to the biological characteristics of Peucedanum praeruptorum Dunn,the field management of Peucedanum praeruptorum Dunn was studied to formulate the field management operation rules.4.Four groups of different planting factors were set,and the biomass,water content,total ash content,alcohol soluble extract,content of Peucedanin A,content of Peucedanin B and inorganic elements were investigated to determine whether the medicinal material-Peucedanum praeruptorum Dunn meet the standards of the national pharmacopoeia,and meawhile,the most suitable planting mode was obtained.Results 1(1)Peucedanum praeruptorum Dunn seed germination were greatly influenced by temperature,and under the condition of 25 ? its germination rate was the highest,up to 46.00%;at 15 ? and 35 ? its germination rate was significantly reduced,only 32.33% and 23.00% respectively;at 5 ? seed germination did not occur.(2)Under different light and dark conditions,the germination rate of Peucedanum praeruptorum Dunn seeds reached about 45%,indicating that the germination of Peucedanum praeruptorum Dunn seeds was less affected by light.(3)Double layer filter paper was more conducive to the germination of seeds,with germination rate up to 36.67%.(4)Different concentrations of GA3,6-ba,2,4-D and KMnO4 all have a certain degree of effect on promoting the germination of Peucedanum praeruptorum Dunn seeds,while at low concentration,it could promote the germination of Peucedanum praeruptorum Dunn seeds,but at high concentration,it could inhibit the germination of Peucedanum praeruptorum Dunn seeds to some extent,and when the concentrations of GA3,6-ba,2,4-D and KMnO4 reached 100mg/L,10mg/L,0.3mg/L and 0.01mg/L respectively,the germination rates were 61.33%,58.67%,55.33% and 55.00%;different concentrations of IAA,ABA and ?-NAA had different degrees of inhibitory effect on the germination rate of Peucedanum praeruptorum Dunn seeds,and the germination rate of the seeds decreased with the increase of the concentration,and the germination rate at the lowest concentration of IAA?ABA??-NAA was 39.67%,45.67% and 47.67%;different concentrations of compound sodium nitrophenolate had different degrees of promoting effect on seeds of Peucedanum praeruptorum Dunn,and this promoting effect was enhanced with the increase of its concentration,and the germination rate was 60.67% at 100mg/L.2.The seeds were divided into three grades according to seed size;for the first grade,germination rate should not be less than 55%,the purity should not be less than 90%,the 1000-seed weight should not be less than 2.5g,the germination potential should not be less than 25%,the water content should not be more than 11%,and the viability should not be less than 95%;for the second grade,germination rate should not be less than 45%,purity should not be less than 85%,1000-seed weight should not be less than 2.0g,germination potential should not be less than 15%,water content should not be more than 14%,vitality should not be less than 90%;the third grade below those percentage.3.The atmosphere,water quality and soil quality of the planting base all met the environmental requirements of Peucedanum praeruptorum Dunn planting;the operating regulations of Peucedanum praeruptorum Dunn field management were formulated.4.The biomass of Peucedanum praeruptorum Dunn treated with residual root,compound fertilizer,25*25cm and sowing in drill were the highest in the four large group treated by four different factors;the effective components of water content,total ash content and alcohol soluble extract basically met Green Industry Standard for Import and Export of Medicinal Plants and Preparations;Praeruptorin A of Peucedanum praeruptorum Dunn treated with the residue root,no fertilization,10*10cm and broadcast sowing was of the highest amount in different groups;Praeruptorin B of Peucedanum praeruptorum Dunn treated with annual growth,organic fertilizer,10*10cm,and sowing in drill was of the highest amount.Conclusion 1.The establishment of seed quality grading standards can regulate the circulation of seeds,ensure seed quality and lay a foundation for standardized planting.2.Temperature and substrate had a relatively large effect on seed germination of Peucedanum praeruptorum Dunn,while light had no significant effect on seed germination;the germination of Peucedanum praeruptorum Dunn seeds was closely related to the species and concentration of exogenous growth regulators.Understanding and mastering the influence of the species and concentration of different plant growth regulators on the germination of Peucedanum praeruptorum Dunn seeds can provide certain theoretical basis for the breeding,the large-scale promotion and cultivation of Peucedanum praeruptorum Dunn.3 The planting base in Linqi town of Chun 'an county meets the requirements of Peucedanum praeruptorum Dunn planting base,and can ensure its output and quality after standard planting.4.After four different groups of Peucedanum praeruptorum Dunn with different factors were investigated,it was suggested to use annual growth factor,organic fertilizer,25*25cm and broadcast sowing for planting.
