This study aims to provide a theoretical basis for the innovation and utilization of summer soybean germplasm by evaluating the main agronomic and economic traits of 262 summer soybean germplasm resources mainly from Sichuan,Chongqing and Jiangsu through field experiments.According to the indoor hydroponic test and field validation,the low-phosphorus-tolerant germplasm Dongdadou-2 and the low-phosphorus-sensitive germplasm Heidou were screened.Preliminary study on the molecular mechanism of soybean tolerance to low phosphorus by RNA-seq technology using these two germplasms as materials.The main findings are as follows:1.The phenotypic diversity of the main agronomic and economic traits of 262 summer soybean germplasm resources was studied.It was found that the summer soybean germplasm resources had very rich phenotypic diversity.Principal component analysis of germplasm resources showed that a total of four principal components were extracted,with a cumulative contribution rate of 80.753%.Based on the main agronomic and economic traits of the tested materials,the squared Euclidean distance method was used to analyze the summer soybean material in the cluster,and the materials were divided into 4 categories and 8 sub-categories.2.Through hydroponic test,17 phosphorus efficiency related traits of summer soybean were measured and analyzed,and the results showed that there were great differences among the traits,and there were significant correlations.Using the principal component analysis factor load matrix,the low phosphorus resistance of the test material was weighted,and the sum of square of deviation was used to cluster the summer soybean materials by using the Euclidean genetic distance,and the material was divided into 4 categories,and 10 genotype of low phosphorus tolerance and 10 genotype of low phosphorus sensitive materials were obtained.The field validation test showed that,Dongdadou-2 was a geno-type of low phosphorus tolerance and Heidou was a genotype of low phosphorus sensitive.3.The expression differences of different genotype soybean under different phosphorus concentration environment were studied by using RNA-seq technique,and the mo-lecular mechanism of soybean resistance to low phosphorus was revealed from the transcription level.It was found that under the condition of low phosphorus,soybean may resist the stress of adverse environment by speeding up the metabolism of cell phospholipids and the synthesis of acidic phosphatase.At the same time,it was found that a gene encoded with purple acidic phosphatase-3 Glyma.05G247800 may be a new gene encoded acidic phosphatase in soybean.
