Soybeans from Hunan and Hubei provinces belong to the southern muti-corpping region. They were valuable accessions of soybean germplasm resources. In this study, 70 Hunan accessions and 206 Hubei accessions which were selected from the primary core collection in China were analyzed by SSR markers and morphological traits. It provided academic evidence to develop and utilize soybean germplasm resources in Hunan and Hubei provinces, and it provided the evidence of three ecotype soybean evolvement. The results are summarized below:1. In this study, similarities and differences of morphological genetic diversity were defined. Through comparison of genetic diversity based on 14 agronomic traits for Hunan and Hubei soybean germplasm, the result indicated that the genetic diversity was similar in qualitative characters but different in quantitative characters. 2. Formula was revised by sampling number during analysis of genetic diversity. Through analysis of sample number, alleles number and Shannon's index, study methods were discussed. Study results advised that formula was amended and it was H=-LnN*. Through comparison of genetic diversity based on 60 SSR markers for Hunan and Hubei soybean germplasm, the result indicated that the genetic diversity of Hunan soybean germplasm was higher than of Hubei.3. This study brought forward the characteristics of distributing and spread. The center of genetic diversity was located in 30~31°N by analysis of SSR markers for Hunan and Hubei soybean germplasm resources. The mode of spared distributed from the center of genetic diversity to south and north. 4. The genetic differentiation between Hunan and Hubei province was less distinct than among different ecotype were found. The result of clustering indicated that the genetic distance was 0.0589 and Fst was 0.0108 between Hunan and Hubei soybean germplasm. Through analysis of genetic relationship for Hunan and Hubei soybean, the result indicated that the relationships between provinces were nearer and the genetic relationships between different ecotype were farer. The result evidenced the rationality which core collection was based on the differentiation among soybean ecotype in China soybean germplasm resources in DNA level.5. Through analysis of genetic diversity, we selected 41 pairs of SSR primers and provide theoretic evidence to study the genetic diversity for Hunan and Hubei soybean.6. The evolvement rule among different ecotype was advanced. The result of cluster analysis showed that genetic relationship of autumn sowing soybean was far from other two ecotypes, because they clustered singly in one group. We speculated that the evolutionary relation is from autumn sowing soybean to the other ecotype soybean and provided molecule evidence for the original cultivated soybean from south region.
