China is the country with the largest number of geese in the world and has a long history.Geese can eat a large amount of pasture and make full use of crude fiber feed.With the development of China's aquaculture industry,vigorously developing and using roughage is an important way to develop the grain-saving livestock husbandry.Corn stalks are by-products of crops with high yield and widespread distribution in Northeast China.Effective development and utilization is an important means to increase economic efficiency.In this study,Jilin white goose was used as the experimental animal,corn straw was used as the source of roughage,and different crude fiber content diets were prepared.Through feeding and slaughtering tests,the methods of histology,biochemistry and molecular biology were used.To study the effect of fiber level on the growth and development of Jilin white geese and the change of cecal microbial diversity,and further explore the law of digestion of crude fiber and the response of microbes in the intestine.A total of 135 healthy Jilin white geese aged 42 days were randomly divided into 3 groups of 45 in each group.Three replicates were set up,each of which had 15 gooses.The crude fiber content of the diet was 6.00%,8.00%,and 10.00%,respectively.,feeding period is 42 days.Through the feeding test,slaughter comparison test,histological section analysis,blood biochemical index analysis,intestinal flora analysis and other test methods,analyze the microbial flora associated with fiber digestion and decomposition.The results show that:1.Diets with different fiber levels had a significant effect on the growth performance of Jilin white goose.There was no significant difference in initial body weight between the three groups of birds with fiber levels of 6%,8%,and 10%.After 42 days of feeding each group of diets,there was a difference in body weight.Significantly,the 6% fiber level group was significantly higher than the 8% fiber level group,and the 8% fiber level group was significantly higher than the 10% fiber level group.The average daily weight gain and feed weight ratio of 6% fiber level group was better than 8% fiber level group,but the difference was not significant,8% fiber level group was better than 10% fiber level group,and the difference was significant.The slaughter rate of 6% fiber level higher than the 10% fiber level group.2.Diets with different fiber levels had significant effects on the blood biochemical indicators of Jilin White Geese,including alkaline phosphatase(ALP)and aspartate aminotransferase(AST).The 8% fiber level group had alkaline phosphatase(ALP)and cereal grass.Transaminase(AST)was significantly higher than 6% fiber level group and 10% fiber level group;The thymus index and the bursal index 6% fiber level group and 8% fiber level group were significantly higher than the 10% fiber level group.3.On the intestinal development of Jilin White Goose.In the histological section data,the data of duodenum,jejunum,and ileum villus heights were the same,and the 8% fiber level group was significantly higher than 6% fiber.Horizontal group and 10% fiber level group.The crypt depth data was significantly higher in the 8% fiber group of the duodenum than in the 6% fiber group;the 6% fiber level group was significantly higher than the 10% fiber level group.The 8% fiber level in the jejunum and ileum intestine was significantly higher than that in the 6% fiber group and the 10% fiber group.Muscle thickness data for the 8% fiber level higher than the 10% fiber level group.4.The main phyla in the caecal contents of Jilin white goose are: Firmicutes,Tenericutes,Actinobacteria,Proteobacteria,Bacteroidetes).The dominant genus is Pediococcus,which can promote fiber digestion and absorption,improve growth performance,promote the growth of beneficial bacteria in the intestine,and inhibit the reproduction of harmful bacteria.There are significant differences among the three groups of Pediococcus at the level of genus,6% group and 8%.The group was significantly higher than the 10% group,which was consistent with the growth performance trend.In summary,the dietary fiber level has an effect on the growth performance of Jilin white goose,the concentrations of ALP and AST in the blood,intestinal development,and the caecal content flora.The diet suitable for Jilin white geese at 6-10 weeks is added.The fiber level is 8%.
