Chicken coccidiosis is a protozoal disease caused mainly by Eimeria tenella,of which the major manifestation is hemafecia.Chicken coccidiosis mainly cause the high incidence and high mortality rate in chickens aged 7-15 days,seriously affecting the economic development of animal husbandry and causing great economic losses to livestock breeding.At present,the prevention and treatment of chicken coccidiosis depends mainly on the chemical pharmaceuticals and vaccine.Poultry products are easily contaminated by chemical pharmaceuticals and vaccine research is still not widely used in intensive farms.Traditional Chinese medicine with low toxicity,low residue and no drug resistance opens new avenues for the treatment of chicken coccidiosis.Coccidiosis was treated with single herbs and compound herbs.The therapeutic effect was evaluated by counting anti-coccidiosis index,detecting immune index and observing pathological changes.Therefore,this study mainly explored the inhibitory effect of different traditional Chinese medicine aqueous extract on chicken coccidiosis oocysts and the therapeutic effect in vivo.First of all,single Chinese medicine aqueous extract agent stop anti-coccidiostat,bleeding dysentery and heat-clearing and detoxifying was choose for test of oocysts in vitro,and he good effect of inhibiting the spore of oocysts was screeed out.Secondly,based on the compatibility principle of traditional Chinese medicine,the Chinese herbal medicine with good effect is composed of three traditional Chinese medicine compounds.Experiments in vitro are conducted to validate the effect of traditional Chinese medicine.In vivo treatment trials are also conducted.Sixty 14-day-old chicks are randomly divided into 7 groups,including in three Chinese medical group,Diclazuril group,Qiujing group,E.tenella infection group and blank control group.Each group of attack on 5×10~4 eggs per mouse.IL-2,IL-6,IL-10,IL-17 and IL-13 in serum and splenic lymphocytes were detected by ELISA and qPCR,as well as the immune antibody SIgA and IgG content.Cecal was select to make pathological sections to observe the performance of inflammation.Finally,treatment effect of the compound Chinese medicine was comprehensively analyzed through experimental data in vitro and vivo.The experimental results showed that the single Chinese herbal medicines Fructus cnidii,Sophora,betel nut,wild chrysanthemum play the best effect,the above drugs as the main drug,combined with stop bleeding dysentery and heat-clearing and detoxifying with adjuvant of traditional Chinese medicine compound.The experimental results of Chinese herbal compound treatment found that the three prescriptions group,the prescription of the second antibody coccidia index was128.75,reached the level of intermediate coccidiosis.ELISA results showed that prescription two SIgA and IgG content compared with EI group was on the rise,and the difference was significant(P>0.05),but there was no significant difference with the Diclazuril group.The results of qPCR and ELISA showed that the expressions of IL-2,IL-6 and IL-17 in serum and lymphocyte of Chinese medicine group,Diclazuril group and Qiujing group were increased,while IL-10 and IL-13 were declined.The effects are most obvious in the second group prescription compared with other two group prescription.Chinese herbal medicine regulate the body's normal immune response by regulating relative balance between the pro-inflammatory cytokines and anti-inflammatory cytokines.Acording with indicators the above test,combined with pathological findings of the cecum,two prescriptions of Chinese medicine group have best effect.
