| Canine pyometra is one of a most common clinical disease of female dogs,it was concealed and can will be life-threatening if not treated promptly.In order to master its epidemiological rules and pathogenesis,effective prevention and treatment of the disease to provide clinical data and a certain theoretical basis.In this study,2013~2017 canine pyometra cases from animal hospital of Xi’an affiliated with Northwest A&F university epidemiological investigation,and the ovary and uterus from sick dogs of canine pyometra in March 2018~October 2018 was collected for HE staining andimmunohistochemicalmethodson thehistologicalchangesand3β-hydroxysteroid dehydrogenase immunohistochemical positive product distribution and expression of research.The results were as follows:1.According to the epidemiological investigation,70.84%of the cases of pyometriosis in dogs aged 6~10 years in Xi’an area,and 51.89%of the cases in small dogs;The incidence was mainly from April to October every year,accounting for 80.2%.The disease types were more open-cervix type,accounting for57.55%.53.78%of the affected dogs had a normal body temperature at first diagnosis.Blood routine examination showed that the mean value of white blood cells(38.89±9.82)×10~9/L,the mean number of neutral cells(26.06±7.89)×10~9/L,and the mean number of lymphocytes(8.46±2.18)×10~9/L,all of which were higher than the normal range.Serum biochemical examination showed that alkaline phosphatase of open-cervix type and closed-cervix type was significantly higher than that of control group(p<0.01).The closed-cervix type of alanine aminotransferase and creatinine was significantly increased(p<0.01),and the open-cervix type was significantly increased(p<0.05).The closed-cervix of urea nitrogen and serum phosphorus was significantly increased(p<0.05).Ultrasound examination showed that endometrial hyperplasia and thickening were the main types of open-cervix type endometrium,and the dark areas were not obvious.In closed-cervix type,most of the affected dogs have thin uterine walls and prominent dark areas of fluid.2.Pathological anatomic changes.The open-cervix type and closed-cervix type ovarian histology change,ovarian cortex thickening,rich in blood vessels of the corpus luteum and corpus luteum cyst occupy the vast majority of ovarian cortex,corpus luteum cyst wall made up of several layers of corpus luteum cells,ovarian cortex without corpus luteum part there was a lot of different stages of atresia follicles,rare original follicle and development of follicles,accompanied by the reproductive epithelium.The main manifestations of the open-cervix type uterus were endometrial gland cystic hyperplasia,with relatively complete hierarchical structure and thickening of muscular layer and endometrial layer.In the closed-cervix type,most of the affected dogs had thin uterine wall,endometrium degeneration,necrosis and shedding,and a small number of dogs had endometrial gland cystic hyperplasia.3.Distribution and expression of 3β-HSD in ovaries and uterus.The 3β-HSD immunoreactive products in ovary were mainly distributed in corpus luteum and follicles and were higher than those in healthy dogs.Luteal expression was the highest in the closed-cervix type,and follicular expression was the highest in the open-cervix type(p<0.01).The expression of 3β-HSD in all layers of uterine horn was higher than that in healthy dogs,mainly distributed in the endometrial epithelium,closed and open type was significantly higher than healthy dogs(p<0.01);closed and open uterine gland The cortex was significantly higher than that of healthy dogs(p<0.05);the expression of myometrium was the highest in the closed type,but the difference was not significant.In conclusion,pyometriosis is a frequently-occurring disease in middle-aged and elderly female dogs over 6 years old,and presents a certain seasonality,which often leads to changes in hematological indexes,liver and kidney dysfunction and changes in related biochemical indexes with more serious damage to the closed-cervix body type.Most of the ovaries had persistent luteal and luteal cysts,most of the uteri had endometrial gland cystic hyperplasia,and the damage of closed-cervix uterine tissue was more serious.The distribution and expression of 3β-HSD in the ovary and uterus suggest that the dysregulation of steroid hormones is closely related to the occurrence and development of this disease. |
