Lilium is one of the family Liliaceae,a perennial herbaceous bulbous flower,with large flowers and bright colors.It is one of the most important cut flowers in t he world.‘Fangio' and ‘Indian aestas' are two varieties of Lilium Asian hybrid.The flowers are crimson and pink,respectively.They have the characteristics of leafless burning,strong disease resistance,long flowering period and high ornamental value.However,the traditional breeding method has a long breeding cycle and a small breeding volume.Bulbils are special reproductive organs of some lily species.After maturity,they can naturally fall off and develop into new intact plants.However,there are very few lily varieties that naturally grow bulbils.Neither ‘ Fangio' nor ‘Indian aestas' can form bulbils naturally.Attempting to induce bulbils formation through in vitro induction culture and decapitation and using a combination of paclobutrazol and chlormequat has important application value for improving the reproductive efficiency of lily.The main findings are as follows:(1)In vitro induction culture of two kinds of lilies.Taking the stem section of a single stem with axillary leaves as explants,the best disinfection method was screened with 1% Na Cl O for 10 min after 75% alcohol disinfection.Both the disinfection effect and the growth status of the explants were the best.The best medium for inducing bulbils is MS + 0.5mg/L6-BA + 0.3mg/L NAA + 90g/L sucrose.The induction rate of ‘Fangio' is 63.33%,and the induction rate of ‘Indian aestas' 76.67%,this medium can induce bulbils directly on the leaf axils of two kinds of lilies,and the diameter of the bulbils is relatively large.The histologically identified in vitro induced bulbils structure is similar to that of Lilium lancifolium,both of which are from the epidermis of the leaf axils.The lower cells begin to divide and gradually form a globular primordium.The induction ability of stem segments of different parts of the same plant of two kinds of lilies was upper > middle > lower.The induction rate of the upper stem segment of ‘Fangio' was 79.17%,and the lower part was 29.17%.The induction rate of upper stem segment of ‘Indian aestas' It is 83.33%,and the lower part is 25%.With the development of the two types of Lilium,the ability of the stem to induce bulbils gradually decreased,the explant induction rate was highest in the vegetative growth stage,and the explant induction ability was the lowest in the late flowering stage.For the subculture of induced bulbils,the optimal proliferation medium was selected as MS + 1.0mg/L 6-BA + 0.2mg/L NAA + 60g/L sucrose with a proliferation coefficient of 3.23.The best method for bulbils rooting is to add 6g/L agar to 1/2 MS medium,the rooting rate is 90%,the average root length is 1.6cm,the average rooting number is 7.6,and the root growth is uniform and good.(2)Field induction was performed on the ‘ Fangio' and ‘Indian aestas' growing in the open field.The best induction method for both was to spray PBZ after removing flower buds.The induction rate of the treated ‘Fangio' was 63.33%.The induction rate was 62.50%.After treatment,the plant leaves increased,stem diameter increased,and pla nt height decreased.The ability to induce bulbils in the three stages is: early budding stage > middle budding stage > initial flowering stage,and the overall induction effect of ‘Fangio' is better than that of ‘Indian aestas'.
