The fruit and leaves of blue honeysuckle are rich in phenolic substances and have certain research value.The fruits and leaves of wild blue honeysuckle collected from 7 areas of the Greater Khingan Mountains?Jagedaqi,Xinlin,Hongwei,Amur,Tuqiang,Tahe,Mohe?and 5 areas of the Lesser Khingan Mountains?Tieli,Cuiluan,Youhao,Wuying,Tangwanghe?,as test materials were compared for the differences of the total phenol content?TPC?and total anthocyanin content?TAC?;DPPH·and ABTS+·free radical scavenging,ferric reduction capacity?FRAP?and oxygen radical absorption capacity?ORAC?analysis to assess antioxidant activity of the crude polyphenol extract of fruit and leaf and its anti-?-amylase,anti-?-glucosidase,anti-lipase activity was also tested.In addition,Solid Phase Extraction Technology?SPE?was used to separate the crude polyphenol extracts of fruits and leaves,and the separated non-anthocyanin polyphenols and anthocyanins were identified with liquid chromatography and mass spectrometry?HPLC-PAD and HPLC-ESI-MS2?,to the explore main biologically active components in fruits and leaves of 12areas,and the differences in the active components between the fruits and leaves of wild blue honeysuckle were compared.It is hoped that providing a theoretical basis for the development and utilization of wild blue honeysuckle in the Greater Khingan Mountains and the Lesser Khingan Mountains.The main findings are as follows:?1?The analysis of the total phenolic content and total anthocyanin content found that the total phenolic content and total anthocyanin content of fruit of wild Lonicera caerulea L.in the Greater Khingan Mountains and the Lesser Khingan Mountains areas were significant difference?P<0.05?.The total phenolic content in leaves(69.91?140.39 mg GAE 100 g-1dw)was significantly higher than that of fruits(31.12?108.52 mg GAE 100 g-1).The average total phenol content of fruits and leaves in the Lesser Khingan Mountains was significantly higher than that in the Greater Khingan Mountains.The total anthocyanin content in the range of 41.26?56.51 mg C3G 100 g-1 dw is also higher than that in the the Greater Khingan Mountains(16.99?52.30 mg C3G 100 g-1 dw).?2?The analysis of antioxidant activity revealed that the fruits and leaves of wild Lonicera caerulea L.in the Greater Khingan Mountains and the Lesser Khingan Mountains have high antioxidant activity.The antioxidant capacity of fruit in different regions of is different.Tangwanghe and Tahe have a strong ability to scavenge DPPH·free radicals.The fruit of Lonicera caerulea L.in Jiagedaqi and Mohe have a good absorption capacity;The oxidation resistance of blades in different areas are similar and different.For example,Jiagedaqi and Youhao not only have a strong ability to remove DPPH·free radicals,but also have a strong ability to reduce ferric iron;Amur has a strong absorption capacity on oxygen free radicals.Through comparison,we also found that the antioxidant capacity of leaves was stronger than that of fruits.?3?The analysis of the inhibition of?-amylase,?-glucosidase,and lipase revealed that both the fruit and leaves of wild Lonicera caerulea L.in the Greater Khingan Mountains and the Lesser Khingan Mountains had inhibitory effects on?-amylase,?-glucosidase,and lipase,but there is a certain difference).The fruit of Youhao has the strongest inhibitory ability,and its IC50 value is only 20.11?g/m L and lower than the IC50 value of acarbose.The leaves of Wuying had the strongest inhibitory ability on?-amylase activity,with IC50 value of 0.29?g/m L.The leaves of had stronger inhibitory activity on?-amylase and?-glucosidase than fruits.with IC50 values of7.84?g/m L,respectively;The fruit of Jiagedaqi and the leaves of Mohe have the strongest inhibitory ability on?-glucosidase activity,the highest inhibition rate of lipase,up to 97.77%,75.70%,respectively.?4?Correlation analysis shows that the fruit of TPC and the antioxidant activity measured by the DPPH method showed a very significant correlation?P<0.01?,and the antioxidant activity measured by the ABTS method showed a significant correlation?P<0.05?,and the leaf of TPC and the anti-oxidant activity measured by the ORAC method showed an extremely significant?P<0.01?.Whether it's fruit or leaves,there is a positive correlation between TPC and anti-?-amylase and anti-?-glucosidase activities,but TAC and anti-?-amylase and anti-?-glucoside had a negative correlation.In addition,there is a positive correlation between TPC and TAC in fruits and leaves and their anti-lipase activity.?5?The identification and analysis of polyphenols in fruits showed that 8 kinds of anthocyanins were detected in fruits of Lonicera caerulea L.in 12 regions;among them,the peak area of cyanidin-3-glucoside is the highest,and their peak areas account for 91%?93%of the total anthocyanin peak area;30 non-anthocyanin polyphenols were also detected in the fruit.Among them,Tuqiang detected the most abundant types of non-anthocyanins;the types of non-anthocyanins in wild blue honeysuckle fruits in the Greater Khingan Mountains were more than those in the Lesser Khingan Mountains;In addition,It is the most that the content of5-caffeoylquinic acid of the wild blue honeysuckle fruits in 12 regions,among which the5-caffeoylquinic acid is the most abundant in Jiagedaqi,accounting for 47%of the total non-anthocyanin polyphenols.?6?The identification and analysis of polyphenols in leaves showed that 1-7 anthocyanins and33 non-anthocyanin polyphenols from the leaves of wild Lonicera caerulea L.in 12 regions are detected.The non-anthocyanin polyphenols in the leaves have three-caffeoylquinic acid,5-p-coumaric quinic acid quercetin and volkensiflavone than the fruit,but no quercetin trihydrate.The non-anthocyanins in the leaves of Tuqiang are also the most abundant,with 19 species.The content of non-anthocyanin polyphenols in the leaves in different regions varies.
