Atractylodes japonica is a traditional Chinese medicine in Northeast China,which has a high medicinal value.In recent years,with the scarcity of its wild resources,Atractylodes japonica is mainly cultivated artificially.However,there are still some problems in the cultivation of Atractylodes japonica,such as low germination rate and difficult emergence.In this paper,the effects of three plant growth regulators(IBA,GA3 and 6-BA)on the seed germination and seedling growth of Atractylodes japonica were studied by means of complete random design,regression orthogonal design and orthogonal design.The regression equation between the three growth regulators and the characteristics of Atractylodes japonica was calculated and the experimental scheme of seed germination and seedling growth was optimized It provides a reference for seed treatment in artificial cultivation of Atractylodes japonica.The results are as follows:1.Low concentration of IBA significantly increased the germination rate,germination potential,seed vigor index,seedling rate,root activity and leaf green content of Atractylodes japonica seeds.The best concentration was 100 mg·L-1.2.The effects of IBA concentration,soaking time and culture temperature on seed germination and seedling growth of Atractylodes japonica were studied,the concentration of IBA has a significant effect on the germination rate of Atractylodes japonica seeds,and the regression equation has a good fitting degree,when the concentration is 50 mg·L-1,the germination rate is the highest;the concentration of IBA and seed soaking time have significant effects on the chlorophyll content of Atractylodes japonica seedlings,the regression equation has a good fitting degree,when the concentration is 50 mg·L-1 and the soaking time is 2 h,the chlorophyll content of seedling is the highest.3.The effects of GA3 concentration,soaking time and culture temperature on seed germination and seedling growth of Atractylodes japonica were studied,the concentration of GA3 and soaking time have significant effects on the germination rate of Atractylodes japonica seeds,the regression equation has a good fitting degree,when the concentration is 50 mg·L-1,soaking time is 2 h,the germination rate is the highest.;the concentration of GA3 has a significant effect on the root activity of Atractylodes japonica seeds,and the regression equation has a good fitting degree,the root activity is the highest when the concentration is 50 mg·L-1.4.The effects of 6-BA concentration,soaking time and culture temperature on the seed germination and seedling growth of Atractylodes japonica were studied,the concentration of 6-BA and soaking time have significant effects on the seed germination rate of Atractylodes japonica,the regression equation has a good fitting degree.The germination rate is the highest when the concentration is 3 mg·L-1 and soaking time is 2 h.The soaking time and culture temperature have significant effects on the emergence rate of Atractylodes japonica,the regression equation has a good fitting degree,when the soaking time is 2 h and the culture temperature is 17?,the emergence rate is the highest.5.The results showed that the optimal combination of seed germination percentage and germination potential,seed vigor,root vigor and seedling height of Atractylodes japonica is IBA2GA326-BA1,(IBA 50 mg·L-1+GA3 50 mg·L-1),and the optimal combination that could effectively increase the main root length of Atractylodes japonica seedlings is IBA1GA326-BA2,(GA3 50 mg·L-1+6-BA 3 mg·L-1),the best combination that can effectively increase the emergence rate and chlorophyll content of Atractylodes japonica is IBA2GA326-BA2,(IBA 50 mg·L-1+GA3 50 mg·L-1+6-BA3 mg·L-1).
